芍药苷对人外周血T淋巴细胞CD25、CD69及NF-κB表达的影响
Effects of Paeoniflorin on Expression of CD25,CD69 and Nuclear Factor κB of Human Peripheral Blood T Lymphocytes
【摘要】 目的研究芍药苷对人外周血T淋巴细胞CD25、CD69及NF-κB表达的影响。方法分离健康人外周血T淋巴细胞,抗人CD3单克隆抗体及抗人CD28单克隆抗体共刺激T淋巴细胞活化,加入不同浓度的芍药苷,流式细胞术检测T淋巴细胞CD25、CD69及NF-κB的表达。结果 50μmol/L芍药苷对活化的T淋巴细胞CD25的表达有抑制作用(P<0.05),100,200μmol/L芍药苷对活化的T淋巴细胞CD25的表达有明显抑制作用(P<0.01);100,200μmol/L芍药苷对活化的T淋巴细胞CD69的表达有明显抑制作用(P<0.01);25,50,100,200μmol/L芍药苷对活化的T淋巴细胞NF-κB的表达有明显抑制作用(P<0.01)。结论芍药苷可呈浓度依赖性抑制T淋巴细胞CD25、CD69及NF-κB的表达。
【Abstract】 Objective To study the effects of paeoniflorin on expression of CD25,CD69 and nuclear factor κB(NF-κB)of human peripheral blood T lymphocytes. Methods Human peripheral blood T lymphocytes were isolated and activated by anti-CD3 mAb plus anti-CD28 mAb and co-incubated with paeoniflorin at various concentrations. The inhibition of paeoniflorin on expression of CD25,CD69 and NF-κB of T lymphocytes was analysed by flow cytometry. Results 50 μmol/L of Paeoniflorin inhibited expression of CD25 of activated human peripheral blood T lymphocytes(P<0.05). 100 μmol/L and 200 μmol/L of Paeoniflorin significantly inhibited expression of CD25 of activated human peripheral blood T lymphocytes(P<0.01). 100 μmol/L and 200 μmol/L of Paeoniflorin significantly inhibited expression of CD69 of activated human peripheral blood T lymphocytes(P <0.01). 25,50,100 and 200 μmol/L of Paeoniflorin significantly inhibited NF-κB of activated human peripheral blood T lymphocytes(P <0.01). Conclusion Paeoniflorin inhibited expression of CD25,CD69 and NF-κB of human peripheral blood T lymphocytes after costimulated by anti-CD3 mAb plus anti-C28 mAb in a dose-dependent manner.
