参麦对大鼠肾缺血再灌注损伤中TLR4/NF-κB信号通路的影响
Effect of Shen Mai Injection on TLR4/NF-κB signal-transduction pathway of renal ischemia reperfusion injury in rats
【摘要】 目的观察参麦注射液(SM)对大鼠肾缺血再灌注损伤TLR4/NF-κB信号通路的影响,探讨参麦对肾脏保护作用的机制。方法建立大鼠肾缺血再灌注损伤模型,随机分为假手术组(Sham组)、缺血再灌注组(IR组)、参麦预处理组(IR+SM组)。HE法观察肾组织病理学形态,自动生化分析仪测定BUN、Cr水平,实时荧光定量PCR法检测肾组织中TLR4和NF-κB p65 mRNA含量,ELISA法检测血浆中ICAM-1和TNF-α表达。结果 IR组较Sham组肾组织病理学明显改变,BUN、Cr水平、TLR4、NF-κB p65 mRNA含量及ICAM-1、TNF-α表达均升高(P<0.05)。与IR组相比,IR+SM组肾组织病理学改变减轻,BUN、Cr水平、TLR4、NF-κB p65 mRNA含量及ICAM-1和TNF-α表达降低(P<0.05)。结论 SM通过抑制TLR4/NF-κB信号通路,下调其下游ICAM-1和TNF-α表达,发挥肾脏保护作用。
【Abstract】 【Objective】To observe the effect of Shen Mai Injection(SM) on the toll-like receptor 4/nuclear factor-kappa B(TLR4/NF-κB) signal-transduction pathway of renal ischemia reperfusion injury in rats,and to discuss the possible the mechanism established.【Methods】The rat model of renal ischemia reperfusion injury was(Sham Group),the ischemia-reperfusion group(IR Group),and the group receiving SM before ischemia-reperfusion(IR+SM Group).The renal pathological changes were observed by light electron microscope.BUN and Cr were determined and compared.The mRNA expression levels of TLR4 and NF-κ B p65 were detected by quantitative real-time PCR. The expressions of intercellular adhesion molecule-1(ICAM-1) and tumor necrosis factor-alpha(TNF-α) were detected with ELISA method.【Results】The renal pathological changes of rats in IR Group were significantly,the expressions of BUN,Cr,TLR4,NF-κ B p65 and ICAM-1,TNF-α were increased remarkably in IR Group,as compared with Sham Group(P <0.05).The renal pathological changes of rats in IR+SM Group was insignificantly,BUN and Cr were lower than those of IR Group,the expression of TLR4 and NF-κ B p65 were decreased,so were the expression of ICAM-1,TNF-α(P <0.05).【Conclusion】The expression of the TLR4/NF-κ B signal-transduction pathway can be decreased by SM,that can be a therapeutic for renal ischemia-reperfusion injury.
