TRAIL协同紫衫醇诱导骨肉瘤细胞凋亡的实验研究

【摘要】 目的研究TRAIL和紫杉醇联合应用对骨肉瘤细胞的抑制作用,寻找骨肉瘤临床治疗的新方案。方法将TRAIL与紫杉醇单独或联合应用于体外培养的骨肉瘤OS-732细胞,采用MTT法测定其对OS-732细胞的抑制率,并于相差显微镜、荧光显微镜下观察细胞形态的改变。结果 50 ng/mL的TRAIL与10μg/mL紫杉醇联合作用于OS-732细胞24 h后,抑制率为47.49%,明显高于单用TRAIL(50 ng/mL)时的9.85%及单用紫杉醇(10μg/mL)时的20.37%(P<0.01)。细胞形态结构均出现凋亡的特征性变化。结论 TRAIL与紫杉醇联合应用可高效杀伤骨肉瘤细胞,这种杀伤效应是通过促进OS732细胞的凋亡来实现的。

【Abstract】 【Objective】 To study the inhibition effect of TRAIL combined with Paclitaxel on the osteosarcoma cell lines so as to explore the new method of the clinical treatment.【Methods】 TRAIL and Paclitaxel were used on the OS-732 cells individually or jointly so as to observe their synergistic effects on the apoptosis of osteosarcoma cells.【Results】 Twenty-four hours after the application of 50 ng/mL TRAIL and 10 μg/mL Paclitaxel jointly on OS-732 cells,The inhibition rate is 47.49% by MTT essay which is significantly higher than the application of 50 ng/mL TRAIL(9.85%) and 10 μg/mL Paclitaxel(20.37%) respectively(P <0.01).Morphological changes observed with inverted phase contrast microscope,fluorescence microscope showed tumorkilling effect of joint application was much stronger than respective application.【Conclusion】 Combination of TRAIL and etoposide is obviously conducive to killing effect of osteosarcoma cells which were achieved by inducing apoptosis.
