目的探讨血管基质片段(vascular stromal fraction,SVF)辅助游离脂肪移植促进其早期存活的相关机制。方法取5例行腹部抽脂手术女性患者自愿捐赠的脂肪组织分离、培养SVF。4~6周龄裸鼠24只(体重15~18 g),将裸鼠背部左、右侧随机分为A、B组(n=24),采用Coleman技术分别于A、B组裸鼠背部皮下移植脂肪0.5 mL和含5×105个SVF的脂肪0.5 mL。移植后1、3、5、7 d分别取6只裸鼠,取出移植脂肪组织,行大体观察、CD31免疫组织化学染色及Western blot检测分析。结果随着移植时间增加,A、B组移植脂肪组织湿重均呈上升趋势,但各时间点A、B组间比较差异均无统计学意义(P>0.05)。移植后1、3 d两组均未见CD31阳性细胞;5、7 d B组CD31阳性细胞数显著高于A组(P<0.05),两组7 d时CD31阳性细胞数显著高于5 d时(P<0.05)。Western blot检测示,两组移植脂肪中VEGF蛋白表达在移植后第3天达峰值,随后开始下降;1、3、5 d B组VEGF蛋白表达明显高于A组(P<0.05)。两组移植脂肪中HGF蛋白表达在移植后前5 d...
