多种氨基酸软胶囊毒理学安全性实验研究

【摘要】 目的评价多种氨基酸软胶囊的毒理安全性。方法采用小鼠急性经口毒性试验、遗传毒性试验(Ames试验、小鼠骨髓微核试验、小鼠精子畸形试验)和大鼠30 d喂养试验对多种氨基酸软胶囊的毒理学安全性进行评价。结果以最大给药剂量(15000 mg/kg·BW)进行小鼠急性经口毒性试验,未见小鼠出现中毒和死亡现象。Ames试验、小鼠骨髓细胞微核试验和小鼠精子畸形试验3项遗传毒性试验结果与阴性对照组比较差异均无统计学意义(P>0.05)。大鼠30 d喂养试验结果显示动物进食量、食物利用率、体重增长、血常规和血生化指标与阴性对照组比较差异均无统计学意义(P>0.05);对大鼠脏器组织进行病理学检查,未观察到多种氨基酸软胶囊对大鼠脏器组织有明显损害作用。结论该多种氨基酸软胶囊在本实验条件下未显示有急性毒性、遗传毒性及亚慢性毒性作用。

【Abstract】 Objective To assess the toxicological security of multi- amino acids soft capsule as health food. Methods Acute oral toxicity test,genetic toxicity tests( Ames test,bone marrow micronucleus test and sperm abnormality test in mice) and rats 30- day feeding test were conducted to evaluate the toxicological safety of multi- amino acids soft capsule. Results No apparent symptom of poisoning or death was observed at the greatest dose of 15000 mg / kg body weight( BW) for the acute oral toxicity test in mice. Results of the three genetic toxicity tests( the Ames test,the mice sperm abnormality test and the bone marrow cell micronucleus test) showed no significant difference between treated groups and control group. There was also no significant difference in food intake,food utilization rate,rat weight gain,blood count and biochemistry index between treatment groups and control group for the 30d- feeding test. No apparent pathological injury to the major organs of rats was observed in the pathological examination of rats’ tissues. Conclusion The multi- amino acids soft capsule was found no acute toxicity,no genetic toxicity and non- sub- chronic toxicity under the current experimental conditions.
