副溶血性弧菌食源性疾病分离株的病原学研究
Etiology research and early warning of Vibrio parahaemolyticus isolates from food- borne disease
【摘要】 目的对杭州市下城区2002年-2012年副溶血性弧菌(Vibrio parahaemolyticus,Vp)食源性疾病分离株进行血清分型、毒力基因和耐药性鉴定,并进行预警分析。方法按照GB/T 4789.7-2008方法确定分离株血清型,采用复合聚合酶链式反应(Polymerase Chain Reaction,PCR)检测直接耐热溶血素毒力基因(tdh)和直接耐热相关溶血素毒力基因(trh),用环介导等温扩增(loop-mediated isothermal amplification,LAMP)技术检测不耐热溶血素毒力基因(tlh),用WHO推荐的K-B纸片法进行药敏试验。结果 358株Vp中215株为O3∶K6血清型,23株为O4∶K8血清型。358株Vp全部携带tlh基因,297株携带tdh基因,1株携带trh基因。358株Vp仅对头孢噻肟、诺氟沙星、复方新诺明3种抗生素100%敏感。结论引起辖区食源性疾病的主要血清型为O3∶K6。tlh基因是共同基因,大多数分离株携带tdh基因。头孢噻肟、诺氟沙星、复方新诺明为病人首选药。
【Abstract】 Objective To understand the serotypes,virulence genes and drug resistance of Vibrio parahaemolyticus isolated from food-borne diseases in Xiacheng district of Hangzhou from 2002 to 2012,so as to provide basis for early warning. Methods In accordance with the test methods specified in GB /T4789. 7-2008,the serotypes of isolates were identified. Polymerase chain reaction( PCR) was employed to detect the direct heat-resistant hemolysin virulence genes( tdh) and related hemolysin virulence genes( trh). Loop-mediated isothermal amplification( LAMP) assay was used to detect the thermolabile hemolysin virulence genes( tlh),and susceptibility test was carried out by K-B paper method recommended by the WHO. Results In 358 Vp strains,215 Vp isolates were O3: K6 serotype,and 23 isolates were O4: K8 serotype. All 358 Vp strains carried tlh gene,297 strains carried tdh gene,and one strain carried the trh gene. The 358 Vp strains were 100% sensitive to cefotaxime,norfloxacin,cotrimoxazole. Conclusion O3: K6 is the main serotype to cause food-borne disease. The tlh gene was common gene of the 358 Vp strains. Most isolates carried tdh gene. Cefotaxime,norfloxacin,cotrimoxazole are the first choice for clinical treatment of food-borne diseases.
