53例羊水细胞ABO血型基因鉴定

【摘要】 目的通过PCR-SSP基因技术检测胎儿羊水细胞ABO血型基因型,产前诊断胎儿ABO血型。方法选取了53例孕16周-25周的孕妇,抽取羊水细胞并进行细胞培养2 d~5 d,然后分离羊水细胞,提取羊水细胞DNA,运用PCR-SSP技术分析其ABO血型基因型;选取出生后的婴儿,采集脐带血或外周血,并用玻片法和微柱凝胶ABO血型卡检测其ABO血型。结果 53例羊水标本中有48例羊水标本通过PCR-SSP方法检测出了ABO血型的基因型;53例孕妇中有24例胎儿的脐带血或外周血经过了血清学和微柱凝胶ABO血型检测,有3例标本未能检测出结果;21例胎儿脐带血或外周血的ABO血型与经基因鉴定的胎儿羊水细胞ABO血型完全一致。结论 PCR-SSP技术可以准确地检测胎儿羊水细胞的ABO血型。

【Abstract】 Objective To analyze the ABO genotype of amniotic fluid cells by polymerase chain reaction with sequence- specific primers( PCR- SSP),so as to accurately identify fetal ABO blood group. Methods Amniotic fluid was collected from 53pregnant women with gestational age of 16 W ~ 25 W for cell culture 2d ~ 5d,then the amniotic fluid cells were separated to extract the DNA. ABO genotypes of amniotic fluid cel1s were detected by polymerase chain reaction with sequence- specific primers. After the fetuses were born,peripheral blood and cord blood was collected from these newborns to detect the ABO genotypes by slide method and micro- column gel agglutination assay. Results ABO genotypes of 48 amniotic fluid specimens were identified in all 53 cases by polymerase chain reaction with sequence- specific primers. While the ABO blood groups of cord blood and peripheral blood from 53 newborn babies were identified in 24 cases by slide method and micro- column gel agglutination assay. In addition to three of those without results,the ABO blood group results of the other 21 newborn babies were the same by the two methods. Conclusion ABO blood group of amniotic fluid cells can be accurately identified by polymerase chain reaction with sequence- specific primers.
