负载bFGF的壳聚糖微球制备及其体外缓释性能测定

【摘要】 目的:制备负载碱性成纤维细胞生长因子(bFGF)的壳聚糖微球,并测其体外释药特性。方法:采用乳化交联法制备壳聚糖空白微球,再进行bFGF吸附,制成负载bFGF的壳聚糖微球,并对微球进行扫描电镜观察。ELISA法测定其载药量与包封率,以及bFGF的体外释放情况。并测定壳聚糖空白微球的吸水膨胀率。结果:负载bFGF的壳聚糖微球直径约4μm,载药量为1.06μg/mg,包封率为84.8%。体外释放实验初期释放bFGF速度较快,随后逐渐趋于平缓。壳聚糖空白微球的吸水膨胀率为650%。结论:采用乳化交联法制备包裹bFGF的壳聚糖缓释微球,简单易行,成球性好,得率高,具有良好的缓释效果。

【Abstract】 Objective:To prepare basic fibroblast growth factor(bFGF) loaded chitosan microspheres,and to analyze the property of controlled release of bFGF in vitro.Methods: Firstly,chitosan blank microspheres were prepared with emulsification cross-linking method,then bFGF-loaded chitosan microspheres were prepared by the chitosan blank microspheres absorbing bFGF.The microspheres were observed by scanning electron microscopy,and the drug loading,encapsulation efficiency and in vitro controlled release of the bFGF-loaded chitosan microspheres were measured by ELISA.Furthermore,the water absorption expansion rate of microspheres were measured.Results: The mean diameter of the bFGF-loaded chitosan microspheres was about 4 μm,and the drug loading and encapsulation efficiency of the bFGF-loaded chitosan microspheres were 1.06 μg/mg and 84.8%,respectively.The microspheres released quickly at initial 12 hours,and then slowed gradually in vitro.The absorbing water expansion rate of the blank microspheres was 650%.Conclusion: Chitosan microspheres encapsulating bFGF prepared by emulsification cross-linking method exhibit high yield and good controlled-release effect.
