超分支滚环扩增技术快速检测番茄溃疡病菌
Rapid detection for Clavibacter michiganensis subsp. michiganensis using Hyperbranched Rolling Circle Amplification
【摘要】 根据番茄溃疡病菌(Clavibacter michiganensis subsp.michiganensis,cmm)的一段特异致病基因pat-1序列,按照锁式探针的设计原理设计探针和扩增引物,优化系列反应条件,建立了特异性的cmm超分支滚环扩增技术。实验结果表明该检测技术能够从供试的10种菌中特异性地检出番茄溃疡病菌,检测灵敏度高,DNA最低检测浓度为500 fg/μL。
【Abstract】 Padlock probe was designed based on the sequence of the unique hypothetic pathogenicity gene pat-1 in complete genome of Clavibacter michiganensis subsp.michiganensis(cmm),according to the design principle of Padlock probe and primer.Detection system of hyperbranched rolling circle amplification(HRCA) was established and optimized.Results show that cmm could be detected specifically by this method,while other reference plant pathogens could not be detected.The detection sensitivity of HRCA for DNA concentration is about 500 fg/μL.
