MSCs对辐射诱导小鼠胸腺瘤中T细胞CD4/CD8各亚群及各亚群中CD45比例变化的影响

【摘要】 目的观察MSCs对辐射诱导小鼠胸腺瘤中T细胞CD4/CD8各亚群及各亚群中CD45比例变化的影响,探讨MSCs对辐射诱发肿瘤的预防和治疗作用。方法应用Kaplan经典方法制备C57BL/6幼年鼠胸腺瘤模型[1],经小鼠尾静脉输注同系鼠MSCs,6个月后取胸腺组织,将胸腺细胞制成单细胞,标记CD4、CD8、CD45,流式细胞术分析T细胞亚群。结果流式细胞术分析结果显示:照射后CD4-CD8-亚群比例由正常组的(5.11±5.10)%降低到(0.01±0.01)%(P<0.05),MSCs治疗(0.61±0.39)%后有所恢复;MSCs治疗组(91.49±4.47)%中CD4+CD8+亚群比例与正常组(82.44±6.06)%接近,单纯照射组(99.09±0.49)%显著高于正常组(P<0.05);照射后CD4+CD8-亚群由(7.72±3.37)%下降到(0.87±0.49)%,CD4-CD8+亚群由(4.74±2.59)%下降到(0.02±0.01)%,MSCs治疗后CD4+CD8-和CD4-CD8+都有不同程度上调。正常组、单纯照射组、MSCs治疗组中CD45+在CD4+CD8+中的比例分别为(86.09±2.24)%、(0.16±0.03)%、(97.35±3.23)%,差异有统计学意义(P<0.05);单纯照射组CD4-CD8-、CD4-CD8+和CD4+CD8-中CD45+均有不同程度的降低,MSCs治疗组有不同程度升高,且具有统计学意义(P<0.05)。结论 MSCs可使辐射诱导胸腺瘤模型组小鼠胸腺中T细胞CD4/CD8各亚群及各亚群中CD45比例增高或恢复,提示MSCs具有防治辐射诱发胸腺瘤的作用,并对辐射造成的胸腺损伤具有修复作用。

【Abstract】 Objective Each CD4/CD8Tcell subsets in the proportion of all cells and CD45in the proportion of each subgroup were observed,to investigate the prevention and treatment of MSCs to radiation-induced thymoma.Methods Kaplan classic method was applied to prepared C57BL/6 young mouse thymoma model[1],and fellow rat’s MSCs was injected via the tail vein,after 6months we get the thymus tissue,then make the thymus cells to single cells and marked CD4,CD8and CD45,and flow cytometry was used to analyze T cell subsets.Results Flow cytometry results showed that after irradiation,CD4-CD8-subsets decreased from the normal group(5.11±5.10)%to(0.01±0.01)%(P<0.05),after the treatment of MSCs(0.61±0.39)%,it has restored;CD4+CD8+subsets in group of MSCs’treatment(91.49±4.47)% was closed to the normal group(82.44±6.06)%,irradiation group(99.09±0.49)% was significantly higher than the normal group(P<0.05);after irradiation,CD4+CD8-subsets decreased from the normal group(7.72±3.37)%to(0.87±0.49)%,CD4-CD8+subsets decreased from(4.74±2.59)%to(0.02±0.01)%,after the treatment of MSCs,CD4+CD8-and CD4-CD8+increased in different degrees.CD45+in the proportion of CD4+CD8+ in normal group、irradiation group and treatment group respectively was(86.09±2.24)%、(0.16±0.03)%、(97.35± 3.23)%,the differences were significant(P<0.05);CD45+in the proportion of CD4-CD8-、CD4-CD8+and CD4+CD8in irradiation group decreased in different degrees,after the treatment of MSCs,they increased in different degrees,the differences were significant(P<0.05).Conclusion MSCs can make each CD4/CD8Tcell subsets in the proportion of all cells and CD45in the proportion of each subgroup increase or restore,it shows that MSCs have prevention and treatment to radiation-induced thymoma,and have repair to the thymus injury that was caused by radiation.
