HPLC测定复脉灵胶囊中乌头碱、新乌头碱、次乌头碱的含量

【摘要】 目的:建立复脉灵胶囊中乌头碱、新乌头碱、次乌头碱的含量测定方法。方法:采用HPLC,流动相甲醇-0.5%的三乙胺(70∶30),Aglient TC-C18色谱柱(4.6 mm×250 mm,5μm),检测波长235 nm,流速1.0 mL·min-1,柱温室温。结果:乌头碱的进样量在0.080 8~0.808 0μg具有良好的线性关系(r=0.999 6),平均回收率98.40%,RSD 2.84%;新乌头碱的进样量在0.866~4.330μg具有良好的线性关系(r=0.999 7),平均回收率98.04%,RSD 2.05%;次乌头碱的进样量在0.089~0.890μg具有良好的线性关系(r=0.999 8),平均回收率98.71%,RSD 2.34%。结论:方法操作简便、灵敏度高、结果准确,可作为复脉灵胶囊的含量测定方法。

【Abstract】 Objective: To establish a method for the determination of aconitine,mesaconitine and hypaconitine in Fumailing Capsules.Method: The aconitine,mesaconitine and hypaconitine in the capsules was determined by HPLC on the Aglient TC-C 18 column(4.6 mm × 250 mm,5 μm) at room temperature.The mobile phase was consisted of methanol-0.05% triethylamine(70 ∶ 30).The flow rate was 1.0 mL ·min-1and the detection wavelength was set at 235 nm.Result: The content of aconitine had a good linearity in the range of 0.080 8-0.808 0 μg(r = 0.999 6),the average recovery was 98.40%,RSD was 2.84%.The content of mesaconitine had a good linearity in the range of 0.866-4.330 μg(r = 0.999 7),the average recovery was 98.04% and RSD was 2.05%.The content of hypaconitine had a good linearity in the range of 0.089-0.890 μg(r =0.999 8),the average recovery was 98.71%,and RSD was 2.34%;Conclusion: This method was simple,reproducible and accurate,and it can be used for determination of Fumailing Capsules.
