基因芯片技术研究环维黄杨星D对大鼠肾脏基因表达的研究

【摘要】 目的:采用基因芯片技术研究环维黄杨星D(CVB-D)对大鼠肾脏基因表达谱的影响,初步探索其对肾脏毒性作用的机制。方法:10只SD大鼠随机分为正常组和CVB-D 10 mg.kg-1组,连续腹腔给药14 d,分别从大鼠肾脏组织中提取总RNA,分离纯化后,反转录合成掺入生物素标记的cDNA探针,与基因芯片杂交,扫描芯片荧光信号图像后,统计处理并进行分析。选取5个差异表达基因,逆转录-聚合酶链反应(RT-PCR)法进行验证。结果:CVB-D组与空白组比较,共筛选出309条差异表达的基因,其中表达上调基因有147条,下调基因有162条,涉及凋亡通路相关基因、细胞周期相关基因、Ca2+信号通路相关基因、MAPK信号级联等。结论:CVB-D的肾毒性可能与凋亡通路、MAPK信号级联、细胞周期及Ca2+信号通路异常调控相关。

【Abstract】 Objective: To investigate the regulated renal gene expression prolife of cyclovirobuxine-D(CVB-D) in rats,and explore its mechanism of renal toxicity.Method: Ten SD rats were divided into 2 group randomly: normal group and CVB-D 10 mg.kg-1group.Rats were continuous gavaged 14 days.The total RNAs were isolated and purified from kidney of normal and CVB-D 10 mg.kg-1rats.Both the mRNAs from two kinds of tissues were reversely transcribed to cDNA with the incorporation of fluorescent labeled hybridization probes.The mixed probes were hybridized to cDNA chip,and then were screened through the analysis of fluorescent intensity.RT-PCR was used to confimed the results of 4 choosing gene chip.Result: Compared with the control group,gene expression of 309 were changed,147 gene expression up-regulated,and 162 gene expression down-regulated.All of the gene changes were related with apoptotic signal,MAP Kinase cascade,calcium-mediated signaling,and cell cycle proteins.Conclusion: The renal toxicity of CVB-D involved in regulating specific genes abnormal expression including apoptosis,MAP Kinase cascade,Calcium-mediated signaling and cell cycle protein,etc.
