正交试验优选刺五加提取物缓释微球的制备工艺

【摘要】 目的:优选刺五加提取物缓释微球的制备工艺。方法:以载药量和包封率的综合评分为指标,采用正交试验法考察海藻酸钠质量分数、刺五加提取物与海藻酸钠的质量比、氯化钙质量分数和交联时间对制备工艺的影响。结果:最佳制备工艺为海藻酸钠质量分数3.5%,刺五加提取物与海藻酸钠的质量比1∶1,氯化钙质量分数4.0%,交联时间2 h,载药量和包封率分别为10.11%,21.74%。制备的刺五加提取物缓释微球呈黑褐色、外观形态较圆整,粒径约0.45 mm,12 h累积释放率达96.12%。结论:优选的制备工艺简单易行。

【Abstract】 Objective:To optimize preparation technology of Acanthopanax senticosus extract sustained-release microsphere.Method:With comprehensive scoring of drug-loading and entrapment efficiency as index,orthogonal test was used to investigate influence of four main factors on preparation technology,including the mass fraction of sodium alginate and CaCl2,A.senticosus extract-sodium alginate and cross-linking time.Result:Optimum Preparation technology was:the mass fraction of sodium alginate 3.5%,A.senticosus extract-sodium alginate 1∶1,cross-linking time 2 h,the mass fraction of CaCl2 4.0%,drug loading and entrapment efficiency of prepared microsphere were10.11%,21.74%,respectively.Prepared A.senticosus extract sustained-release microsphere was dark brown and roundness,particle size was about 0.45 mm,cumulative release was up to 96.12% in 12 h.Conclusion:Optimized preparation technology was simple and reliable.
