SPA预处理对小鼠金葡菌致死性攻击的保护作用

【摘要】 目的:探讨金葡菌A蛋白(SPA)预处理小鼠对金葡菌致死性攻击的保护作用。方法:于0、24小时对BALB/c小鼠行腹腔注射SPA 20μg/只/次,对照组以生理盐水替代SPA。第二次处理24小时后,腹腔注射致死量(3×1010CFU/kg)金葡菌(ATCC25923),观察小鼠生存率;非致死量金葡菌(2×108CFU/只)腹腔感染6小时后,ELISA、Q-PCR检测小鼠血清及脾脏中TNF-α、IL-6、IFN-γ含量及表达量。结果:与对照组小鼠2天内全部死亡比较,致死量细菌攻击后,SPA预处理组小鼠2周生存率为30%,显著高于对照组(P<0.05);非致死量细菌感染后,预处理组小鼠血清及脾脏中TNF-α、IL-6、IFN-γ含量及表达量显著低于对照组(P<0.05),IL-10表达量降低。结论:SPA预处理增强BALB/c小鼠抵御S.aureus致死攻击。

【Abstract】 Objective: To study the protective effect of SPA pretreatment on mice challenged with lethal-dose Staphylococcus aureus.Methods: BALB/c mice were given intraperitoneal injections(i.p) of 20 μg SPA or normal saline at 0 h and 24 h on consecutive 2 days.All of mice were then challenged by an i.p injection of live Sthaphylococcus aureus(ATCC25923) with lethal-dose(3×1010 CFU/kg) at 1 day after the second pretreatment.The mice mortality was observed,and the serum level of IFN-γ,TNF-α and IL-6 in mice infected with non-lethal live Staphylococcus aureus(2×108 CFU for each,i.p) were detected by ELISA.Meanwhile,the gene relative expression of these cytokines in splenocyte was detected by Q-PCR.Results:Compared with normal saline treated,the pretreated with SPA prolonged significantly the survival of mice challenged with lethal live S.aureus(P<0.05).The level and gene relative expression of TNF-α,IL-6,IFN-γ and IL-10 of serum and splenocyte in mice that were administrated with SPA were reduced(P<0.05).Conclusion:Pretreatment with SPA protects the mice from challenge with lethal and non-lethal live S.aureues.
