柴胡与4种伪品的DNA指纹研究

【摘要】 目的建立中药柴胡及其常见伪品数字指纹,并用于柴胡鉴别研究.方法采用改良CTAB法、SDS法和改良SDS法提取柴胡及其伪品基因组DNA,随机引物PCR扩增,琼脂糖凝胶电泳分离PCR扩增产物,对琼脂糖凝胶电泳结果进行数字处理.结果改良SDS法提取DNA纯度优于SDS法和改良CTAB法;琼脂糖凝胶图谱转化成数字指纹.结论运用改良SDS法能够提取到纯度较高的基因组DNA,数字指纹更直观、易懂.

【Abstract】 Objective To establish the DNA fingerprint of bupleurum and its counterfeits and use it for the identification of bupleurum.Method The improved CTAB method,SDS method and the improved SDS method were applied to extract cDNA of bupleurum and its imitations,the random primers was amplified with PCR,agarose gel electrophoresis was used to isolate the products of PCR amplification,and finally,the results were analyzed in a digital processing way.Results The purity of DNA obtained with the improved SDS method was better than that with the improved CTAB method,and the atlas of agarose gel was translated into digital fingerprint.Conclusion The application of improved SDS method can extract DNA with higher purity of genome.The digital fingerprint is more intuitive and easy to be understood.
