α-硫辛酸对2型糖尿病大鼠肝脏脂质沉积的改善作用

【摘要】 目的探讨α-硫辛酸(α-lipoic acid,ALA)对高脂联合链脲佐菌素(STZ)诱导的SD大鼠肝脏脂质沉积的影响。方法健康雄性8周龄SD大鼠45只,随机分为正常对照组15只,造模组30只,造模组应用高糖高脂饮食加30 mg/kg STZ诱发2型糖尿病模型,成模大鼠再随机分成非药物干预糖尿病组(DM组,n=15)和α-硫辛酸干预组(ALA组,60 mg/kg,n=15),干预8周。干预结束后留取血清和肝脏组织,行生化及氧化应激指标检测,组织形态学检测,并应用Western blot的方法测定大鼠肝脏组织脂代谢相关蛋白固醇调节元件结合蛋白-1(sterol regulatory element binding protein-1,SREBP-1)的表达。结果造模组大鼠血清TG[(1.6±0.69)mmol/L]和TC[(3.14±1.1)mmol/L]含量明显升高。ALA治疗后,血清TG[(0.69±0.13)mmol/L]和TC[(1.67±1.0)mmol/L]明显降低,肝功能和氧化应激得到了改善,肝脏中的脂质沉积明显减少。SREBP-1的蛋白表达水平明显减低。结论 ALA干预可改善高脂饮食联合STZ诱导的SD大鼠的肝脏脂质沉积,其机制可能与改善氧化应激及减少SREBP-1的表达有关。

【Abstract】 Objective In this study, we addressed the issues by examining the effects of α-lipoic acid(ALA) treatment for hepatic steatosis in SD rat fed by High-Fat combined with STZ. Methods Male 8 weeks SD rat were from Animal centre, Guangdong Medical College. All rats were randomly divided into two groups: SD +chow diet group and SD+High Fat group for 8 weeks. After that, in order to get the diabetes model rats, the high fat group rat were injected with STZ(30 mg/kg). Then all rats were randomly divided into two groups: DM+ saline group, DM+ALA group for 8 weeks. Systemic evaluations were followed with:Body weight and FBG were measured every 2 weeks. Triglycerides(TG), total serum cholesterol(TC) were measured. Hematoxylin-eosin staining(HE) and Oil-red-O staining for liver. Protein expression of Sterol regulatory element binding protein-1(SREBP-1) was tested by Western Blot. Results The results showed ALA statistically decreased the body weight,(370.2±30.5)g vs.(509.6±47.8)g, TG(0.69±0.13)mmol/L vs.(1.6±0.69)mmol/L, TC(1.67±1.03)mmol/L vs.(14±1.1)mmol/L and oxidative stress. Significant reduction of hepatic lipid accumulation was observed according to the histology results. Meanwhile, ALA statistically decreased the protein expression SREBP-1. Conclusion ALA ameliorated hepatic steatosis probably though reduced lipogenesis and oxidative stress in DM rat.
