重组人血管内皮抑素对H22腹水瘤小鼠腹膜血管电镜形态影响的研究
Study on effect of recombinant human endostatin on H22 ascites tumor vascular morphology of mouse peritoneum
【摘要】 目的观察重组人血管内皮抑素(Endostar,YH-16,恩度)对H22腹水瘤小鼠腹膜血管超微电镜结构形态的影响,为Endostar治疗恶性腹腔积液提供病理形态学依据,进一步探讨恶性腹腔积液形成的机制。方法应用H22肝癌细胞系建立小鼠腹水瘤模型,40只健康BALB/C小鼠随机分为4组,即0.9%生理盐水对照组、低剂量Endostar组(5mg/kg)、中剂量Endostar组(10mg/kg)和高剂量Endostar组(15mg/kg)。小鼠腹腔积液长出后隔日腹腔注射实验药物共3次,停药1d后每天随机处死小鼠2只/组,取出H22腹水瘤小鼠腹膜,利用透射电镜技术,观察各组小鼠腹膜血管超微结构形态学改变。结果电镜下:与生理盐水组相比,3个剂量Endostar组可见毛细血管壁增厚,部分趋于正常,毛细血管通透性降低;血管内皮细胞核较小,核内异染色质增多;细胞间有紧密连接;细胞内线粒体水肿明显减轻,并可见较多正常线粒体。结论应用Endostar后,H22腹水瘤小鼠腹膜血管超微结构会发生形态学改变,此种改变为Endostar治疗恶性腹腔积液提供了病理形态学依据。
【Abstract】 Objective To observe the effect of recombinant human endostatin on H22 ascites tumor in mice peritoneal ultrastructure of vascular.To provide morphological basis support for endostar in treating malignant ascites.Further discussion on the mechanism of formation of malignant ascites.Methods H22 hepatoma cell lines establish ascites tumor model.40 healthy BALB/C mice,after intraperitoneal inoculation,were randomly divided into four groups:saline negative control group;Endostar low group(5 mg/kg);Endostar medium group(10 mg/kg);Endostar high group(15 mg/kg).When the peritoneal effusions grow,intraperitoneal injection of the experimental drug for 3 times.killed mices and removed peritoneals,morphologic alterations of the peritoneal was examined by transmission electron microscopy.Results Compared with normal saline,Endostar 3 dose groups visible the capillary wall thickening or part of normal,then capillary permeability in reducing;Vascular endothelial cell nucleus smaller nuclei heterochromatin increased;Intercellular tight junction;Intracellular mitochondrial swelling significantly reduce and more become normal.Conclusion After apply Endostar,H22 peritoneal ascites tumor in mice vascular ultrastructural morphological change can occur,such changes as Endostar in treating malignant seroperitoneum provide a morphological basis.
