艾灸神阙穴对肾阳虚家兔肾组织超氧化物歧化酶基因表达的影响
Effects of Moxibustion on expression of SOD of rabbit with kidney-yang deficiency
【摘要】 目的探讨艾灸神阙穴对衰老的影响和作用机制。方法将48只家兔随机分成8组,即Ⅰ组(空白对照组)、Ⅱ组(模型组)、Ⅲ组(神阙预防组)、Ⅳ组(神阙加足三里预防组)、Ⅴ组(神阙加命门预防组)、Ⅵ组(神阙治疗组)、Ⅶ组(神阙加足三里治疗组)、Ⅷ组(神阙加命门治疗组),除空白组外,均采用羟基脲片灌胃法造成肾阳虚家兔模型。三个预防组灌胃的同时分别施以艾炷灸,三个治疗组在造模成功后分别予以艾炷灸治疗,观察各组家兔肾组织超氧化物歧化酶(SOD)mRNA的表达。并通过单用神阙穴与神阙穴加足三里、神阙穴加命门等穴位组合进行比较,探讨不同穴位配伍对以上指标的影响。结果Ⅲ、Ⅳ、Ⅴ造模结束后出现少许肾阳虚症状(P>0.05),经艾灸治疗后肾组织中Cu/Zn-SOD mRNA和MnSOD mRNA的相对表达量明显高于Ⅱ组(P<0.01),且较Ⅰ组无显著差异(P>0.05);造模后Ⅵ、Ⅶ、Ⅷ组出现明显的肾阳虚症状(P<0.01),经艾灸治疗后,症状明显减轻(P<0.01),且肾组织中Cu/Zn-SOD mRNA和Mn-SOD mRNA的相对表达量也明显高于Ⅱ组(P<0.01),而与Ⅰ组比较,差异不明显(P>0.05);艾灸神阙、神阙加足三里、神阙加命门三种方法组间比较差异不明显(P>0.05)。结论艾灸神阙、神阙加足三里、神阙加命门三种方法均可有效提高肾阳虚家兔SOD表达量,对肾阳虚家兔具有明显预防及治疗作用。艾灸神阙穴延缓衰老机制与调节自由基代谢关系密切。
【Abstract】 Objective To explore the influence of moxibustion on Shenque( RN8) point on aging and the mechanism. Methods 48 health adult rabbits were randomly divided into 8 groups,a normal group,a model group of kidney-yang deficiency,a prevention group with moxibustion on Shenque( RN8) point,a prevention group with moxibustion both on Shenque( RN8) and Zusanli( ST 36) points,a prevention group with moxibustion both on Shenque( RN8) and Mingmen( GV4) points,a treatment group with moxibustion on Shenque( RN8) point, a treatment group with moxibustion both on Shenque( RN8) and Zusanli( ST 36) points and a treatment group with moxibustion both on Shenque( RN8) and Mingmen( GV4) point. In addition to the normal group,the rabbits poured hydroxyurea. The three prevention groups were treated with moxa cone moxibustion after being poured hydroxyurea. After the success in modeling,the three treatment groups were treated with moxa cone moxibustion. Then detect gene expression of superoxide dismutase( SOD mRNA) in rabbit kidney,compare the effect of the moxibustion from both prevention and treatment aspects. And by comparing the effect of different combination of points,explore their differences on the impact of these indicators. Results A few syndromes of kidney-yang deficiency were found in groupⅢ,Ⅳ,Ⅴ( P > 0. 05),and relative expression of Cu / Zn-SOD mRNA and Mn-SOD mRNA in kidney significantly higher than groupⅡ after the moxibustion treatment( P <0. 01); Compared with groupⅠ,there was no significant difference( P > 0. 05). Obvious symptoms of kidney-yang deficiency were found in groupⅥ,Ⅶ,Ⅷ( P < 0. 01),although the syndromes of kidney-yang deficiency were improved and relative expression of Cu / Zn-SOD mRNA and Mn-SOD mRNA significantly higher than groupⅡ after the moxibustion treatment( P < 0. 01); And no remarkable difference was found between groupⅠ and them( P > 0. 05). There was no significant difference between the three combination of points( P > 0. 05). Conclusions Three moxibustion treatment can all enhance the expression of SOD mRNA of Rabbit with Kidney-yang deficiency effectively,and have the significant effect both at prevention and treatment aspects. But there was no significant difference between the three moxibustion treatments,which may be related to the specificity of Shenque. Whereas the mechanism of moxibustion on Shenque point on aging may be related to regulating the free radical metabolism.
