VP16/三元LDH复合材料的制备表征及性质

【摘要】 依托泊苷(VP16)是一种广谱性抗癌药物。分别采用共沉淀和离子交换法制备了VP16/三元LDH纳米载药复合材料,以提高药物的抗肿瘤性能,降低药物的生物毒性。研究结果显示,VP16/三元层状氢氧化物纳米复合载药体系呈片层结构,所得纳米粒分布均匀,其中由共沉淀法和离子交换法(即经氨基酸预插层)制备的谷胱甘肽/层状双氢氧化物纳米复合体系的载药量分别为13.69%和20.36%,具有良好的缓释性能。细胞毒性实验结果证明了以三元层状氢氧化物材料作为依托泊苷载体,可降低依托泊苷的细胞毒性,且纳米载药复合体系对肿瘤细胞活性抑制率达60%,远大于药物本身的抑制率32%。结果表明LDH纳米材料作为药物分子VP16的载体,能在提高肿瘤抑制效果的同时降低一定的正常细胞毒性作用,并且实现药物的定量控释传递,可作为一种新型的抗肿瘤复合片剂。

【Abstract】 To improve the anticancer efficiency and decrease toxicity,the anticancer drug etoposide(VP16) has been encapsulated in the galleries of Mg-Zn-Al LDH by coprecipitation and ion exchange methods.The results suggested that the VP16/LDH was well suspended and the drug loadings were 13.96% and 20.36%,respectively.According to the toxicity test results,LDH is bio-compatible.The anticancer efficiency test revealed that the VP16-LDH nanohybrids had higher tumor suppression efficiency(60%) compared to VP16 itself(32%),which is undoubtedly due to the drug delivery ability of LDH.In a word,LDH could be designed as a carrier for VP16 given its better anticancer effect and lower toxicity compared to plain drug.LDH could also achieve controlled release and has potential to be a novel anticancer drug delivery system.
