幽门螺杆菌体外培养影响因素探讨

【摘要】 目的探索影响幽门螺杆菌体外培养的影响因素。方法观察不同培养时间幽门螺杆菌的菌落特点和细菌形态特点,确定幽门螺杆菌的最佳培养时间;通过SYTO9/PI核酸染料对暴露于空气中不同时间的幽门螺杆菌进行死活菌的染色,估计幽门螺杆菌在空气中的存活时间。结果培养3d的幽门螺杆菌菌落为针尖大小、形态为典型弯曲杆菌,培养超过3d的幽门螺杆菌菌落直径可达1mm,菌体形态多为球型;荧光染色检查显示,暴露在空气中的幽门螺杆菌随着暴露时间的延长逐渐死亡,4h后几乎全部死亡。结论幽门螺杆菌体外培养最佳培养时间为3d,且菌体形态典型。幽门螺杆菌暴露于空气中的存活时间≤4h。

【Abstract】 Objective To explore factors affecting the in vitro culture of Helicobacter pylori.Methods Colonies and the bacterial morphology of H.pylori were observed at different times.Bacterial nucleic acids were stained with fluorescent SYTO9/PI to ascertain appropriate culturing times and estimate survival time after exposure to air.Results The bacterial colonies were as small as the tip of a needle and the bacteria had a typical spiral/corkscrew morphology.After culturing for 3 days,the colonies enlarged to 1 mm in size and the bacteria had a spherical morphology.Fluorescence staining indicated the H.pylori were dead after exposure to air for 4 hours.Conclusion The optimal culturing time for H.pylori is 3 days,and the resulting bacteria will have a typical morphology.Helicobacter pylori survives for less than 4 hours when exposed to air.
