青蒿素对秀丽隐杆线虫寿命和生殖的影响

【摘要】 目的用不同浓度青蒿素处理秀丽隐杆线虫,观察秀丽隐杆线虫的生存周期、繁殖及相关基因的变化,探讨青蒿素影响秀丽隐杆线虫生殖的机理。方法用不同浓度青蒿素处理线虫,在体视显微镜下观察线虫的存活天数以及产卵情况。提取各组线虫RNA,反转录成cDNA后进行实时荧光定量PCR,比较sec-23、lin-29、ung-1表达的差异性。结果 3、4和5mmol/L青蒿素组线虫和正常对照组的平均生存天数分别为3.65、3.25、2.65和4.3d,差异有统计学意义(t=0.021,P<0.05);3、4和5mmol/L青蒿素组线虫均未产卵或后代出现;5mmol/L青蒿素组线虫sec-23表达下调,lin-29与ung-1表达均上调。结论青蒿素能缩短秀丽隐杆线虫的寿命,抑制其生殖能力,作用机理可能与sec-23、lin-29和ung-1的调控有关。

【Abstract】 Objectives To explore the effects of artemisinin on the life cycle and reproduction of Caenorhabditis elegans and changes in levels of expression of related genes and to discuss the mechanism of artemisinin’s effectiveness.MethodsC.elegans served as a model organism.Different concentrations of artemisinin were added to NGM plates and C.elegans was cultured for about 24 hours.The number of living C.elegans was counted and plates were checked for eggs daily.Total RNA of C.elegans was isolated and transcribed into cDNA.Real-time RT-PCR was used to compare levels of expression of sec-23,lin-29,and ung-1.Results C.elegans lived an average of 3.65 days with a drug concentration of 3 mmol/L,3.25 days with a concentration of 4 mmol/L,and 2.65 days with a concentration of 5 mmol/L while the control lived an average of 4.3 days.The difference in survival time statistically significant(t=0.021,P<0.05).No eggs or offspring were noted after treatment with artemisinin at a concentration of 3,4,and 5 mmol/L.The expression of sec-23 was up-regulated and the expression of lin-29 and ung-1 was down-regulated with an artemisinin concentration of 5 mmol/L.Conclusion Artemisinin reduces the lifespan and reproduction of C.elegans.The mechanism of artemisinin’s effectiveness might involve sec-23,lin-29,or ung-1.
