人血浆中匹维溴铵的LC-MS/MS法测定

【摘要】 建立了液相色谱-串联质谱法测定人血浆中的匹维溴铵。以紫杉醇为内标,采用C18色谱柱,流动相为乙腈-2 mmol/L乙酸铵溶液-甲酸(70:30:0.1),多反应监测(MRM)扫描,监测离子对m/z 512.5→231.3(匹维溴铵)和m/z 854.3→286.3(紫杉醇)。低、中、高浓度匹维溴铵的血浆提取回收率分别为111.7%、104.9%和99.7%,日内和日间RSD均小于15%。

【Abstract】 A LC-MS/MS method was established for the determination of pinaverium in human plasma.Paclitaxel was used as the internal standard.A C18 column was used with the mobile phase of acetonitrile-2 mmol/L ammonium acetate-formic acid(70:30:0.1) in multiple reaction monitoring(MRM) mode with the transitions of m/z 512.5→231.3(pinaverium) and m/z 854.3→286.3(paclitaxel),respectively.The recoveries of pinaverium at low,medium and high concentration levels were 111.7%,104.9% and 99.7%,respectively.The inter-and intra-day RSDs were less than 15%.
