盐酸金刚烷胺的合成
Synthesis of Amantadine Hydrochloride
【摘要】 用金刚烷与乙腈、20%发烟硫酸"一锅法"反应制得1-乙酰胺基金刚烷,再经氢氧化钠水解、与盐酸成盐制得抗病毒药盐酸金刚烷胺,总收率约82%。
【Abstract】 Adamantane reacted with acetonitrile in 20% fuming sulfuric acid to get N-(1-adamantyl)acetamide,followed by hydrolysis with sodium hydroxide to obtain amandine,the latter was converted to antiviral drug amantandine hydrochloride by salification with hydrochloric acid.The overall yield was about 82%.
