液-质联用测定人血浆中己烯雌酚浓度的不确定度分析

【摘要】 目的:研究高效液相色谱-串联质谱法(HPLC-MS/MS)测定人血浆中己烯雌酚浓度的不确定度评定方法。方法:对HPLC-MS/MS法测定血浆中己烯雌酚浓度的全过程进行分析,确定不确定度来源,最后根据各分量计算出合成不确定度并进行了扩展。结果:置信概率为95%时,血浆低(0.20ng.mL-1)、中(1.96ng.mL-1)、高(50.41ng.mL-1)质量浓度己烯雌酚的扩展不确定度分别为0.16、0.18、2.48ng.mL-1。结论:本法适用于评价HPLC-MS/MS法测定血浆中己烯雌酚浓度的不确定度研究,对于血药浓度的测定具有重要参考意义。

【Abstract】 OBJECTIVE To develop a method for evaluation the uncertainty in the determination of diethylstilbestrol in human plasma by high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS).METHODS The content of diethylstilbestrol in human plasma was determined by HPLC-MS/MS.Uncertainty sources were obtained from analysis of the whole procedure of HPLC-MS/MS method,and then,the combined standard uncertainty and expanded standard uncertainty were calculated accordingly.RESULTS The expanded uncertainty for low,medium and high level of diethylstilbestrol was 0.16,0.18 and 2.48 ng·mL-1,respectively(P=95%).CONCLUSION This method is suitable for the uncertainty evaluation of diethylstilbestrol determination in human plasma by HPLC-MS/MS.The establishment of the method is of reference significance to the detection of plasma concentration.
