近红外光谱快速测定柴胡总皂苷肠溶片包衣膜厚度研究

【摘要】 目的利用近红外光谱分析技术,建立柴胡总皂苷肠溶片包衣厚度快速、无损测定方法。方法对光谱的预处理方法、建模波段、主因子数的选择进行详细地讨论,比较不同建模方法,并对模型进行方法学考察。结果采用偏最小二乘回归方法(PLSR)建立包衣膜厚度的近红外光谱定量校正模型。所建模型的相关系数为r=0.991 5,内部交叉验证均方根误差(RMSECV)为3.6μm,预测均方根误差(RMSEP)为3.24μm。结论预测结果表明,本实验所建方法快速、无损、可靠,可应用于中药生产包衣过程的在线检测。

【Abstract】 OBJECTIVE To establish a rapid and nondestructive method to determine coating thickness of the total saponin of Radix Bupleuri enteric coated tablets by near infrared spectroscopy( NIRS) analytical technique. METHODS Spectral pretreating methods,wavenumber selection and factors of NIRS were discussed in detail. Different modeling methods were compared and the methodology of model was investigated. RESULTS Partial least squares regression( PLSR) was used for building the quantitative calibration model. Correlation coefficients( r),root-mean-squares error of cross-validation( RMSECV),and root mean square error of prediction( RMSEP) obtained by PLSR model were 0. 991 5,3. 6 and 3. 24 μm respectively. CONCLUSION Predicted results show that the established method is rapid,nondestructive,and reliable,which can be applied to the online monitor of Chinese medicine tablets coating process.
