微透析技术研究左氧氟沙星在大鼠脑和血中的药动学
Pharmacokinetics of Levofloxacin in Rat Brain and Bloods
【摘要】 目的研究左氧氟沙星静脉给药后游离型药物在大鼠血和脑中的药动学。方法采用微透析技术对大鼠脑组织和血液进行同步取样,联合反相高效液相色谱法检测脑组织和血液中盐酸左氧氟沙星的浓度,用WinNonlin软件处理药动学参数。结果大鼠血液中的游离左氧氟沙星浓度在给药后10 min时最高,脑组织在20 min到达峰浓度,然后持续下降;前4 h,血液中游离药物浓度高于脑组织中相应时间点药物浓度,之后两种组织中游离药物浓度水平相当。血液、脑组织的药时曲线下面积分别为(21.17±7.83)和(7.00±2.65)μg·h·mL-1。结论微透析技术联合反相高效液相色谱检测方法可以成功用于左氧氟沙星在脑组织和血液中的药动学研究。
【Abstract】 OBJECTIVE To investigate the pharmacokinetics of levofloxacin in rats brain tissues and blood.METHODS Brain tissues and blood were sampled by microdialysis simultaneously.The concentrations of levofloxacin in both blood and brain tissues were measured by high performance liquid chromatography.The concentration-time data was analyzed by WinNonlin software.RESULTS The maximum concentrations of free levofloxacin in blood and brain tissues were reached at 10 min and 20 min,respectively,then both continuously decreased.The concentration of free levofloxacin in blood was higher than that in brain tissues in the first 4 h,then returned to similar leve1.The area under the curve(AUC) of free levofloxacin was(21.17±7.83) μg·h·mL-1 in blood and(7.00±2.65) μg·h·mL-1 in brain tissues.CONCLUSION Microdialysis with reversed phase high performance liquid chromatography established in this study can be used to determine the pharmacokinetics of levofloxacin in brain tissues and blood.
