RP-HPLC法测定罗布麻叶饮片中金丝桃苷的含量

【摘要】 目的:建立测定罗布麻叶饮片中金丝桃苷含量的方法。方法:采用反相高效液相色谱法。色谱柱为Phenomenex Luna C18(250mm×4.6mm,5μm),流动相为乙腈-0.015mol/L磷酸二氢钾溶液(1∶9,V/V,用氢氧化钾或磷酸调pH=5.0),流速为1.0ml/min,柱温为30℃,检测波长为260nm。结果:金丝桃苷的质量浓度在10~200μg/ml范围内与其峰面积积分值呈良好的线性关系(r=0.9998);精密度、稳定性、重复性试验的RSD均<2%;平均加样回收率为97.8%,RSD=1.5%(n=9)。结论:该方法简单、可行,结果准确、重复性好,可用于罗布麻叶饮片的质量控制。

【Abstract】 OBJECTIVE:To establish a method for the content determination of hyperoside in Apocynum venetum decoction pieces. METHODS:RP-HPLC method was adopted. The determination was performed on Phenomenex Luna C18 (250 mm×4.6 mm, 5 μm)column with mobile phase consisted of acetonitrile-0.015 mol/L potassium dihydrogen phosphate solution(1∶9,V/V,pH adjusted to 5.0 with KOH or phosphoric acid) at the flow rate of 1.0 ml/min. The column temperature was 30 ℃ and detection wavelength was 260 nm. RESULTS:The linear range of hyperoside were 10-200 μg/ml(r=0.999 8)with average recovery of 97.8% (RSD=1.5%,n=9). RSDs of precision,stability and reproducibility tests were all lower than 2%. CONCLUSIONS:This method is simple,feasible,accurate and reproducible. It can be used for the quality control of A. venetum decoction pieces.
