仙珍骨宝对骨质疏松模型大鼠的保护作用研究
Preventive Effect of Xianzhen Gubao on Osteoporosis Model Rats
【摘要】 目的:研究仙珍骨宝对骨质疏松模型大鼠的保护作用。方法:通过灌胃醋酸泼尼松(3.5mg/kg)每天1次、连续12周以复制大鼠腰椎骨质疏松模型。实验分为正常对照(等容生理盐水)、模型(等容生理盐水)与仙珍骨宝高、低剂量(0.95、0.475g/kg)组。灌胃给药,每天1次,连续12周。末次给药后取大鼠腰椎骨分别测定骨密度、骨生物力学参数和骨形态计量学参数。结果:与正常对照组比较,模型组大鼠腰椎骨密度和断裂载荷显著下降,骨组织总面积、骨小梁面积、骨小梁周长、骨矿化率和生长板荧光标记间距显著减少(P<0.01或P<0.05)。与模型组比较,仙珍骨宝高剂量组大鼠腰椎骨密度、生物力学参数、骨组织总面积、骨小梁面积、骨小梁周长、骨矿化率和生长板荧光标记间距均显著增加(P<0.01或P<0.05)。结论:仙珍骨宝对6月龄SD大鼠灌胃醋酸泼尼松所致的骨质疏松有较好的预防作用。
【Abstract】 OBJECTIVE:To study preventive effect of Xianzhen gubao on posteoporosis model rats. METHODS:Bone loss model of lumbar vertebrae was induced by intragastric administration of prednisone(3.5 mg/kg),All of them were treated intragastrically once a day for 12 weeks.. Model rats were divided into 4 groups,i.e. normal control group(constant volume of normal saline),model group(constant volume of normal saline)and Xianzhen gubao high-dose and low-dose groups(0.95 g/kg,0.475 g/ kg). All of them were treated intragastrically once a day for 12 weeks. After last medication,BMD,biomechanical properties and bone histomorphometric parameters of lumbar vertebrae were determined. RESULTS:Compared with normal control group,BMD, biomechanical properties and histomorphometric parameters(TAr,TbAr,TbPm,MAR)and the interlabeled width of growth plate of lumbar vertebrae were significantly decreased in model group(P<0.05). Compared with model group,BMD,biomechanical properties,bone histomorphometric parameters and the interlabeled width of growth plate of lumbar vertebrae were increased significantly in Xianzhen gubao high-dose group(P<0.05). CONCLUSIONS:Xianzhen gubao has good preventive effect on prednisone-induced osteoporosis in 6 month old SD rats.
