不同比例配伍的延胡索-川楝子药对中延胡索乙素的含量测定

【摘要】 目的:建立测定不同比例配伍的延胡索-川楝子药对中延胡索乙素含量的方法,揭示不同比例配伍的药对的合理性。方法:采用高效液相色谱法。色谱柱为PhenomenexC18(250mm×4.6mm,5μm),流动相为甲醇-0.1%磷酸水(32∶68,V/V),检测波长为282nm,流速为1.0ml/min,柱温为25℃。结果:延胡索乙素的质量浓度在0.023~0.276mg/ml范围内与峰面积积分值呈良好线性关系(r=0.9996);精密度、稳定性、重复性试验的RSD均<2%;平均加样回收率为99.49%,RSD=2.34%(n=6)。延胡索乙素的质量分数随着药对中川楝子用量增加而增加,延胡索和川楝子比例为1∶0→2∶1→1∶1→1∶2时,延胡索乙素质量分数分别增加了0.002%、0.024%、0.008%。结论:该方法重复性好、灵敏度高、简便、准确,可作为延胡索-川楝子药对中延胡索乙素含量的测定方法。

【Abstract】 OBJECTIVE:To establish a method for the content determination of tetrahydropalmatine in different proportion of Corydalis yanhusuo-Melia toosendan,and to reveal the rationality of different proportion of the drug-couple. METHODS:HPLC method was adopted. The determination was performed on Phenomenex C 18 (250 mm×4.6 mm,5 μm)column with mobile phase consisted of methanol-0.1% phosphoric acid(32 ∶ 68,V/V)at the flow rate of 1.0 ml/min. The detection wavelength was set at 282 nm and column temperature was 25 ℃. RESULTS:The linear range of tetrahydropalmatine were 0.023-0.276 mg/ml(r=0.999 6) with an average recovery of 99.49%(RSD=2.34%,n=6). The RSDs of precision,stability and reproducibility tests were all lower than 2%. The contents of tetrahydropalmatine increased with the increase of drug-couple. The proportion of C. yanhusuo to M. toosendan were 1∶0,2∶1,1∶1 and 1∶2,and the contents of tetrahydropalmatine increased by 0.002%,0.024% and 0.008%,respectively. CONCLUSIONS:The method is good in repeatability,high in sensitivity,simple and accurate in operation,and it is suitable for the content determination of tetrahydropualmatine of drug-couple C. yanhusuo-M. toosendan.
