马流感病毒H3亚型血凝抑制试验抗原的研究

【摘要】 为研制马流感病毒(EIV)H3亚型血凝抑制(HI)试验抗原,本研究以华北地区分离的H3N8亚型EIV(A/Equine/Huabei/1/2007(H3N8))为种毒,经条件优化确定了其生产工艺。检测结果表明:制备的EIV H3亚型HI试验抗原与英国兽医参考实验室(VLA)生产的抗原具有同样的敏感性,对EIV感染马后9 d~10 d的血清可检测出抗体阳性;对马流感H3亚型灭活疫苗免疫后31 d的马血清检测抗体为阳性;检测美国肯塔基大学马病研究室提供的强阳性、中阳性和弱阳性血清以及阴性血清的效价,结果与瓶签标定的效价一致。而且,制备的抗原仅与EIV H3亚型阳性血清发生特异性反应,与EIV H7亚型、猪流感病毒H1亚型、猪流感病毒H3亚型等12种其他亚型的A型流感病毒阳性血清均不发生反应。研究结果显示,制备的抗原具有很好的敏感性和特异性,填补了国内空白,可以用于目前EIV H3亚型HI抗体的检测,为马流感的诊断和监测提供了有效的技术手段。

【Abstract】 To prepare the antigen of hemagglutinin-inhibition(HI) test for detection of the H3 subtype equine influenza virus(EIV), the H3N8 EIV A/equine/Huabei/1/2007 was used as stock virus to prepare the EIV H3 subtype antigen for HI test. Under the optimized conditions for the antigen production technology, the results demonstrated that the prepared antigen has the same sensitivity as the antigen produced from the Veterinary Laboratories Agency(VLA) of England, which was able to detect positively with the equine serum at 9 to 10 days post infection with the H3N8 EIV and 31 days post immunization with H3N8 EIV inactivated vaccine, and the same titer was detected with the standard H3 subtype sera of the high, medium, low positive serum and negative serum as the labels provided by Kentuky Universary. In addition, it reacted only with the positive serum of H3 EIV and no cross-reactions with other related standard positive serum of twelve subtypes of influenza A virus, such as positive sera of EIV H7, swine influenza virus H1/H3 subtype, and so on. The study showed that the prepared antigen was sensitivity, specificity and equivent to the foreign products, which provided the bases for application as a commercial reagent for the equine influenza diagnosis, monitoring and quarantine.
