猪肽聚糖识别蛋白pPGLYRP1和pPGLYRP2基因的组织表达规律和品种差异研究
Expression Profiles and Breed Differences of pPGLYRP1 and pPGLYRP2 Gene
【摘要】 本实验旨在研究猪体内2种肽聚糖识别蛋白(PGRPs)pPGLYRP1和pPGLYRP2基因在不同组织的表达规律,并比较两基因在不同品种猪体内的表达差异,为揭示猪PGRPs的表达特点和不同猪种抗病力差异机制提供理论依据。实验选取30日龄莱芜猪和长白猪各6头,屠宰后取肝脏、骨髓、肺、淋巴结、胸腺、脾脏、十二指肠、空肠、回肠,应用实时荧光定量PCR技术,分别检测pPGLYRP1和pPGLYRP2基因在各组织中的表达量。结果表明:两猪种体内pPGLYRP1 mRNA均在骨髓表达量最高(P<0.01),肺次之,其他组织相对低表达;两猪种体内,pPGLYRP2 mRNA均在肝脏表达量最高(P<0.01),小肠次之,其他组织相对低表达;在pPGLYRP1相对高表达的骨髓和肺中,长白猪pPGLYRP1 mRNA表达水平极显著高于莱芜猪(P<0.01);在pPGLYRP2相对高表达的肝脏、小肠中,长白猪pPGLYRP2 mRNA表达水平极显著高于莱芜猪(P<0.01)。
【Abstract】 This study was conducted to investigate ge ne expression profiles of two PGRPs(pPGLYRP1 and pPGLYRP2) in pigs,and to compare the different gene expression of pPGLYRP1 and pPGLYRP2 between Laiwu and Landrace pigs.Six Laiwu and Landrace Pigs of 30 days of age were selected and slaughtered.Tissue samples of liver,bone marrow,lung,mesenteric lymphnodes,thymus,spleen,duodenum,jejunum and ileum were analyszed for gene expression levels of pPGLYRP1 and pPGLYRP2 by real-time PCR.The results showed as follows,in both breeds,pPGLYRP1 gene expressed highest in bone marrow(P<0.01),and expressed high in lung,however it expressed very low in other tissues; in both breeds,pPGLYRP2 gene expressed highest in liver(P <0.01),and expressed high in intestinum tenue,however it expressed very low in other tissues; in bone marrow and lung,pPGLYRP1 gene of Landrace pigs expressed significantly higher than that of Laiwu pigs(P <0.01); in liver and intestinum tenue,pPGLYRP2 gene of Landrace pigs expressed significantly higher than that of Laiwu pigs(P<0.01).
