番茄红素对心肌细胞缺氧损伤的作用机制

【摘要】 目的探讨番茄红素对心肌细胞沉默信息调节因子1(Sirt1)-自噬通路的作用。方法原代培养乳鼠心肌细胞予以无血清、无糖培养及缺氧干预,模拟心肌缺血,用番茄红素和Sirt1的抑制剂尼克酰胺干预。采用Western blot法检测Sirt1、自噬相关基因6(Atg6)Beclin1及自噬相关基因8LC3蛋白的表达;RT-PCR技术检测心肌细胞Sirt1的mRNA表达。结果①在心肌缺氧中,番茄红素提高了细胞Sirt1蛋白和mRNA的表达水平;②番茄红素增加了心肌缺氧时自噬相关基因Beclin1和LC3蛋白的表达;③Sirt1的抑制剂尼克酰胺阻断了番茄红素对自噬蛋白的作用。结论番茄红素可能通过Sirt1激活自噬,从而减少心肌缺氧损伤。

【Abstract】 Objective To investigate the effect of lycopene on Sirt1-Autophagy pathway in cardiomyocytes underhy-poxic condition. Methods The primarily cultured neonatal rat cardiomyocytes were incubated under the condition ofhypoxia in glucose free plus serum deprivation DMEM and exposed to lycopene or/and NAM. The protein level of Sirt1,Atg6(Beclin1) and autophagic marker, LC3 were tested by Western blotting.The mRNA of Sirt1 was detected by Real time PCR. Results Lycopene increased the protein and mRNA expression of Sirt1, and up regulated expression of Beclin1 and LC3 protein underhypoxic condition. Blocking Sirt1 by NAM could significantly decrease the effect of lycopene on autophagy in cardio- myocytes underhypoxic condition.Conclusion Lycopene may reduce myocardial damage by enhancing Sirt1-autophagy pathway underhypoxic condition.
