大鼠肝脏损伤早期NF-κB时序性表达的实验性研究

【摘要】 目的研究大鼠肝脏损伤后细胞核因子-κB(nuclear factor kappa B,NF-κB)在肝脏组织中的表达变化,探讨其表达变化与损伤时间的关系。方法采用自由落体撞击法建立大鼠肝脏损伤模型,应用免疫组织化学方法检测伤后不同时间NF-κB的表达水平,并应用图像分析系统进行分析。结果正常组织中有少量的NF-κB阳性表达。NF-κB在伤后0.5h开始逐渐增高,8h达高峰,12~24h略有下降,但仍保持较高水平。统计分析结果提示:各实验组与对照组及与相邻上组比较差异均有统计学意义(P<0.05)。结论大鼠肝脏损伤后NF-κB表达持续增加,8h达峰值,12~24h持续高表达并出现下降趋势,其规律性表达可望用于肝脏早期损伤时间的推断。

【Abstract】 Objective To study the expression changes of nuclear factor kappa B (NF-κB) in the liver tissues after liver injury of rats and discuss the relationship between expression changes and injury time. Methods Establishment of rat liver injury model by weight-dropping method, the expression levels of NF κB in varied time after injury were tested by immunohistochemistry and were analyzed by image analysis system. Results There were a handful of NF-κB positive expressions in normal tissues. The NF-κB levels gradually increased after 0.5 h of liver injury, reached up to peak in 8 h, and slightly dropped from 12 h to 24 h but still maintained a high level. The statistical analysis displayed differences among the experimental group, control group, and adjacent groups ( P < 0.05). Conclusion After liver injury of rats, the NF-κB expressions first continue to increase, come to peak at 8h, and maintain a high level but with a declining trend from 12 h to 24 h. The regularity expression of NF-κB can be used for injury time deduction of liver at early stage.
