米非司酮对小鼠子宫CD4~+/CD8~+T淋巴细胞及巨噬细胞的影响
Effects of mifepristone on CD4~+/CD8~+ T cells and macrophages in the uterus tissues of mice
【摘要】 为探讨米非司酮(RU486)对小鼠子宫组织中CD4+、CD8+T淋巴细胞以及F4/80+巨噬细胞数量变化和分布的影响,并探讨米非司酮致流产的细胞免疫机制,对怀孕至第7天的小鼠颈部注射米非司酮,并在小鼠怀孕至第8天时采集子宫,应用SP法对小鼠子宫CD4+、CD8+T淋巴细胞及F4/80+巨噬细胞进行免疫组织化学染色,观察并统计一定面积内免疫细胞的数量及其分布。结果显示,RU486诱导流产组的流产率为100%,显著高于对照组的20%(P<0.01)。由RU486诱导的小鼠子宫内CD4+T淋巴细胞的数量显著增加,CD8+T淋巴细胞的数量也有一定的增加,但CD4+/CD8+比值明显升高;流产小鼠子宫内巨噬细胞的数量显著增加,对应的试验小鼠流产率显著升高。结果表明,米非司酮诱导小鼠流产与子宫细胞免疫增强有关。
【Abstract】 In order to investigate the possible cellular mechanism in mifepristone(RU486) induced abortion mice this study was carried out to focus on influences of RU486 on the distribution of CD4+ T cells,CD8+ T cells and F4/80+ macrophages in the uterus tissues. RU486 was given via subcutaneous injection to induce abortion in pregnant mice on day 7 of gestation.All mice were sacrificed on day 8 of pregnancy to collect the uterus samples. The CD4+ T cells,CD8+ T cells and F4/80+ macrophages were tested by immunohistochemistry after incubation with rat anti-mouse CD4+,CD8+,and F4/80+ monoclonal antibodies.The RU486 induced group showed 100% abortion,significantly higher than the control group(abortion rate 20%,P<0.01).The results proved that CD4+ cells in the mouse uterus of the RU486 induced abortion group were significantly increased. The CD8+ T cells were also increased,with the ratio of CD4+/CD8+ being increased sharply(P<0.01).The number of macrophages in the uterus tissues of abortion mice was increased significantly,and the mice abortion ratio was increased too.The study strongly suggests the close relationship of RU486 induced abortion and the enhancement in immunocytes in the uterus.
