绵羊伪狂犬病病毒的鉴定及序列分析

【摘要】 为确诊疑似绵羊伪狂犬病、分析其病原gC基因序列特征,以山东某羊场疑似伪狂犬病病料为对象,经负染后进行电镜观察,PCR扩增特异性gC基因并对其进行序列测定及遗传进化关系分析。电镜下观察可见病毒样粒子,病料中扩增出特异性目的基因片段,序列测定结果显示扩增的目的基因为伪狂犬病病毒(PRV)gC基因片段。遗传进化分析表明该病毒和1998年分离于南美洲阿根廷的猪伪狂犬病病毒遗传关系最近,同属于一个进化分支。流行病学及临床症状和实验室检测结果表明该病例为绵羊伪狂犬病病毒引起,为国内进行羊PRV分子流行病学研究积累了资料。

【Abstract】 To diagnose a suspected sheep pseudorabies case and analyse the sequence of gC gene of sheep pseudorabies virus(PRV),the brain of sheep with clinical symptom of sheep pseudorabies from a farm in Shandong Province was collected in 2012. PRV in the brain tissue was confirmed by observation under electron microscope and PCR amplification and sequence analysis of the gC gene.Phylogenetic analysis was made based on the gC gene.The results confirmed a PRV case,in which the isolate was named as China/SD/2012.Phylogenetic analysis demonstrated that there was a close genetic relationship between the PRV isolate and other PRVs which isolated from Argentina pigs in 1998.This study provided an useful information for the molecular epidemiology of sheep PRV in China.
