应用黑曲霉代谢苊醌的影响因素研究
Influencing factors in biodegradation reaction of acenaphthenequinone by aspergillus niger
【摘要】 采用黑曲霉生物降解苊醌,将苊醌代谢为毒性较低的物质.探讨了反应时间、底物浓度、菌体浓度、pH值和温度对降解率的影响,并设计正交实验优化了降解条件.结果表明:在100mLpH=7.2的缓冲溶液中,摇床反应96h、苊醌浓度为2.74mmol/L、1g(干重)黑曲霉的条件下,苊醌的降解率最高,为93.7%,其中底物浓度对降解率的影响最大.黑曲霉对苊醌的代谢主要为还原反应,通过探讨反应路径,发现控制反应条件可获得较高的手性醇类化合物的收率.本研究为微生物催化还原刚性的前手性酮制备手性醇提供一种新的思路.
【Abstract】 Acenaphthenequinone was detoxicated by the biodegradation of Aspergillus niger.Here five parameters,including reaction time,substrate,strain concentration,pH and temperature were investigated.The results show that the conversion of acenaph thenequinone was 93.7% under optimal conditions.The specified condition were as followings: buffer pH 7.2,2.74 mmol/L substrate,31 ℃,wet strains(dry weight 1 g) and shaking reaction time 96 h.The concentration of substrate was the main reason affecting the percent conversion.The noteworthy pathway of metabolization was bioreduction.Chiral diols could be obtained with high yield by determining the reacting conditions.This method puts forward a new route in preparation of chiral alcohols via reducing polycyclic aromatic ketones.
