陕西省健康儿童肠道病毒的分离与定型
A Study on the Type Characteristic of Enterovirus Isolated from Healthy Children in Shaanxi Province
【摘要】 目的对陕西省健康儿童肠道病毒(Enterovirus,EV)感染情况进行分析,为EV所引起疾病的预防与控制提供参考。方法按世界卫生组织《脊髓灰质炎实验室操作手册》规程进行病毒分离,用逆转录-聚合酶链反应扩增分离到的病毒VP1编码区基因片段,并进行核苷酸序列测定和分析,以鉴定EV的型别。结果对健康儿童159份粪便标本进行病毒分离,其中34份病毒分离阳性,阳性率21.38%。在阳性标本中,埃柯病毒(Enteric Cytopathogenic Human Orphan Virus,ECHO;引起细胞病变的人肠道孤儿病毒)7型(ECHO7)11份,占32.35%;柯萨奇病毒(Coxsackie Virus,CV)B组5型(CVB5)5份,占14.71%;ECHO14、CVB3各3份,各占8.82%;CVA92份,占5.88%;ECHO30、ECHO13各1份,各占2.94%;另外8株病毒未能确定型别。结论陕西省健康儿童中ECHO7感染率最高,且不同地区所分离的EV型别不同,增加了对EV所引起的疾病进行预防与控制的难度。
【Abstract】 Objective To analysis the infection rate of enterovirus among the healthy children under five years old in Shaanxi province,to supply scientific strategies for diseases control and prevention.Methods Isolated virus according to the World Health Organization ( WHO ) developed Polio Laboratory Manual,nucleotide sequences of enteroviruses VP 1 regions were successfully amplified by reverse transcriptase-polymerase chain reaction ( RT-PCR ),RT-PCR products were sequenced and analysised to identify the type of enteroviruses.Results Among the 159 stool specimens collected from four different regions of Shaanxi province,there are 34 positive by virus isolation,isolation rate was 11.3 % .Among the 34 positive samples,there are 11 strains of enteric cytopathogenic human orphan ( ECHO ) virus type 7 ( ECHO7 ),accounting for 32.35 % ;5 strains of coxsackie virus B5 ( CVB5 ),accounting for 14.71 % ;3 strains of ECHO14,accounting for 8.82 % ;3 strains of CVB3,accounting for 8.82 % ;2 strains of CVA9,accounting for 5.88 % ;1 strains of ECHO30,accounting for 2.94 % ;1 strains of ECHO13,accounting for 2.94 % .Conclusions The results showed that the infection of ECHO is highest in shaanxi province in healthy children,and different regions of the isolated intestinal virus type is different.That increase the difficulty in prevention and control of disease which is caused by intestinal virus.
