融合蛋白XE-TNFαm2对SIV的抑制作用
Inhibitory effect of XE-TNFαm2 on SIV-infected cells
【摘要】 目的检测XE-TNFαm2靶向融合蛋白作为新型生物性药物抑制SIV病毒感染CEMx-174细胞的作用。方法 XE-TNFαm2、SIV和CEMx-174细胞共同培养,在培养的第5天,用酶联免疫吸附法(ELISA)测定培养上清中P27抗原水平,间接免疫荧光法(IFA)和流式细胞仪测试SIV阳性细胞,应用PE-annexin V凋亡检测试剂盒测试细胞存活率和凋亡率。结果 XE-TNFαm2在细胞安全最大药物浓度50μg/mL时,对SIV病毒抑制率为40%。细胞早期凋亡率在XE-TNFαm2浓度6.25μg/mL时为37.1%。结论融合蛋白XE-TNFαm2对SIV病毒有一定的抑制作用,其中部分作用推测是因通过诱导感染细胞产生凋亡导致病毒死亡。
【Abstract】 Objective To detect the inhibitory effect of XE-TNFαm2 on SIV infection and apoptosis in SIV-infected cells in vitro.Methods A series of 10-fold dilutions of XE-TNFαm2 was used in the experiment.SIVmac251 and CEMx-174 cells were cultured at 37℃ for 5 days.Anti-SIV activity was detected with P27 enzyme-linked immunosorbent assay(ELISA).The apoptosis rate was analyzed by flow cytometry.Results XE-TNFαm2 showed low cytotoxicity to CEMx-174 cells,and inhibited the cell infection of SIV.The highest inhibition rate was 40%.Conclusion Our results demonstrate that XE-TNFαm2 can inhibit SIV infection and conduct apoptosis in infected cells.
