不同浓度七氟醚对人鼻咽癌细胞株CNE2黏附能力以及CD133表达的影响
Effects of different concentrations of sevoflurane on adhesion and expression of CD133 in human nasopharyngeal carcinoma cell line CNE2
【摘要】 目的观察不同浓度七氟醚对人鼻咽癌细胞株CNE2黏附能力、CD133表达的影响。方法分为四组:对照组(Control组)、1.7%七氟醚组(Treat1组)、3.4%七氟醚组(Treat2组)和5.1%七氟醚组(Treat3组)。Treat1组、Treat2组和Treat3组人鼻咽癌CNE2细胞,经1.7%、3.4%、5.1%七氟醚作用2 h、4 h、6 h。检测细胞黏附率;采用qRT-PCR法检测CD133的mRNA表达。结果与处理前比较,Treat1组、Treat2组和Treat3组处理2 h、4 h和6 h时细胞黏附率降低,CD133的mRNA及其蛋白表达下调(P<0.05)。结论七氟醚可降低人鼻咽癌细胞株CNE2的黏附能力,且呈浓度和时间依赖性,其机制可能与下调CD133的表达有关。
【Abstract】 Objective To investigate the effects of different concentrations of sevoflurane on adhesion and expression of CD133 in human nasopharyngeal carcinoma cell line CNE2. Methods The cells were randomly divided into 4 groups:control group and 3 sevoflurane groups for 2,4 and 6 h respectively. Cell adhesion rate was detected by adhesion test and the expression of CD133 mRNA and protein was determined by qRT-PCR. Results Sevoflurane significantly inhibited the cell adhesion rate and down-regulated CD133 expression in a concentration and duration of expo-sure-dependent manner. Conclusion Sevoflurane can inhibit cell adhesion through down-regulation of CD133 expres-sion.
