参苓白术散对利血平所致脾虚小鼠血清蛋白质组影响

【摘要】 目的用双向电泳技术分析参苓白术散(人参、茯苓、白术等)对利血平所致脾虚证模型小鼠血清蛋白质组的影响,探讨其治疗脾虚泄泻的作用机制。方法采用利血平单因素造模法建立脾虚证小鼠动物模型。分别收集正常组、模型组、高剂量组和低剂量组的血清样本,对样本处理后分别进行双向凝胶电泳,并利用PDQuest8.0图像分析软件对2-DE图谱进行差异性分析。选取差异明显的蛋白点进行基质辅助激光解吸电离飞行时间质谱鉴定,检索数据库鉴定蛋白质。结果经过比较分析四组血清电泳图谱,有明显差异的蛋白点有35个,质谱鉴定出6种蛋白,分别为β球蛋白、维生素D结合蛋白、妊娠区带蛋白、载脂蛋白A-I、载脂蛋白A-IV和白蛋白。结论免疫功能降低、小肠功能紊乱可能是利血平致小鼠脾虚发生的主要病理环节,参苓白术散能直接调节提高免疫功能、促进小肠吸收功能的相关蛋白的表达,使其归于正常,这可能是参苓白术散治疗利血平所致小鼠脾虚的主要作用机制。

【Abstract】 AIM To use two dimensional electrophoreses to analyze the effect of Shenling Baizhu Powder( Ginseng Radix et Rhizome,Poria,Atractylodis macrocephalae Rhizoma,etc.) on mice with diarrhea of spleen deficiency.METHODS Mice model for spleen deficiency were established by administration of Reserpine. The serum samples of normal group,model group,high dosage group and the low dosage Shenling Baizhu Powder group were collected.PDQuest8. 0 image analysis software was used to analyze the differences in two-dimensional gel electrophoresis. Matrix assisted laser desorption ionization. Time of flight mass spectrometry was used for serum proteome. RESULTS There were thirty-five protein spots with obvious differences,and six proteins were identified as beta-globin,vitamin D-binding protein,pregnancy zone protein,apolipoprotein A-I,apolipoprotein A-IV and albumin. CONCLUSION Shenling Baizhu Powder can directly enhance immune function,promote protein expression relative to intestinal absorption,and make mice with spleen deficiency caused by Reserpine return to normal.
