白木香叶与沉香形成相关的信号分子研究

【摘要】 目的应用气相色谱-质谱连用技术结合数据分析对未结香和已结香的白木香树的叶中挥发性成分进行比较。方法 15批白木香(树龄10个月至35年)用三氯甲烷冷浸后超声提取,GC-MS检测,主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)对结果进行判别分析。结果白木香叶中共鉴定出75种挥发性成分,PCA和OPLS-DA进行分类,再用变量重要性投影法挑选得9个对差异贡献最大的潜在标志物;结香的白木香叶中芳香族的量显著降低(P<0.05),萜类成分角鲨烯的量显著升高(P<0.05)。生育酚仅在已结香的白木香叶中检测到。结论未结香和已结香白木香叶挥发性成分存在显著差异,为快速的、非破坏性的沉香药材采收时间及质量评价提供参考依据。

【Abstract】 AIM To distinguish the special volatile constituents of leaves of Aquilaria sinensis with resin from that without resin by GC-MS combined with data analysis. METHODS The volatile components were extracted from fifteen batches of eaglewood leaves by means of chloroform-maceration at the room temperature and ultrasonic extraction detected by GC-MS.The obtained results were analyzed by principal component analysis(PCA) and orthogonal partial least squares discriminant analysis(OPLS-DA). RESULTS Seventy-five kinds of chemical components were identified and classified by PCA and OPLS-DA.Nine components were selected as the potential markers with the larger contribution by variable importance in the projection.In the leaves of heartwood with resin,aromatics contents decreased significantly(P< 0.05),while contents of terpenes(squalene) increased significantly(P< 0.05).Tocopherols,including γ-tocopherol,β-tocopherol and α-tocopherol,were only detected in the leaves heartwood with resin. CONCLUSION There are significant differences between the leaves of Aquilaria sinensis with and without resin.The developed method is rapid,non-destructive to make the reasonable picking time of Aquilaria sinensis.
