楂芪降糖汤对糖尿病肾病大鼠肾脏TGF-β1及VEGF表达的影响

【摘要】 目的研究楂芪降糖汤(山楂、黄芪、粉萆薢、玉米须、葛根、丹参、淫羊藿、玄参等)对糖尿病肾病大鼠肾脏TGF-β1及VEGF表达的影响,探讨其作用机制。方法链脲佐菌素诱导建立糖尿病肾病模型,优降糖作为对照组。成模后各组按相应药物剂量灌胃。治疗10周后,测生化指标;观察肾组织病理形态改变;免疫组化观察肾组织TGF-β1、VEGF的表达。结果各治疗组生化指标变化均优于模型组,且较模型组TGF-β1、VEGF表达下调,尤以楂芪汤高剂量组改善较显著(P﹤0.05)。结论楂芪降糖汤对糖尿病肾病大鼠肾损害有保护作用,其机制可能是通过降低TGF-β1、VEGF表达实现的。

【Abstract】 AIM To investigate the effects of Zhaqi Jiangtang Decoction(Crataegi Fructus,Astragali Radix,Dioscoreae hypoglaucae Rhizoma,Maydis Stigma,Puerariae lobatae Radix,Salviae miltiorrhizae Radix et Rhizoma,Epimedii Folium,Scrophulariae Radix,etc.) on the expressions of transforming growth factor-β1(TGF-β1) and vascular endothelial growth factor(VEGF) in kidney of diabetic nephropathy rats.METHODS The model rats of diabetic nephropathy were established by injecting streptozotosin.After the model was formed,the relevant drugs were given intragastrically to different groups respectively.A group was medicated with glibenclamide as the control.After 10 weeks of treatment,the serum levels of blood urea nitrogen(BUN),creatinine(Scr) for treated diabetic nephropathy rats were detected.The pathological changes of renal tissue were observed by HE staining.The expressions of TGF-β1 and VEGF were determined by immunohistochemical.RESULTS Changes in biochemical indicators in the treatment groups were better than those in the model group.Compared with the model group,the downregulation of TGF-β1,VEGF was obvious in high-dose of Zhaqi Jiangtang Decoction group(P < 0.05).CONCLUSION Zhaqi Jiangtang Decoction has protective effect to diabetic nephropathy rat with kidney damage,and the mechanism may be related to reduction in TGF-β1 and VEGF expressions.
