【摘要】 目的观察通络益肾方对高糖培养下大鼠肾小球系膜细胞增殖、细胞外基质调控系统MMP-9/TIMP-1、TGF-β1表达的影响,探讨其防治糖尿病肾病的可能机制。方法以体外培养的大鼠肾小球系膜细胞为研究对象,将培养的肾小球系膜细胞分为空白对照组、高糖培养组(高糖组)、通络益肾方高、中、低剂量组及西药对照组,用大鼠含药血清给药,分别给予相应处理48 h后,采用四甲偶氮唑盐(MTT)法检测细胞增殖,ELISA法检测细胞培养上清液TGF-β1、MMP-9和TIMP-1的水平。结果与空白对照组比较,高糖组肾小球系膜细胞增殖明显(P<0.01),上清液中TGF-β1和TIMP-1水平显著增加,MMP-9水平减少(均P<0.01);通络益肾方高、中、低剂量均能明显抑制肾小球系膜细胞增殖(P<0.01或P<0.05),下调TGF-β1和TIMP-1表达,上调MMP-9表达(P<0.05或P<0.01),尤以高剂量组效果最佳,呈剂量依赖性;西药对照组上述指标表达水平和中药中剂量组相当(P>0.05)。结论通络益肾方可能通过抑制肾小球系膜细胞增殖和下调TGF-β1、促进细胞外基质的降解而拮抗糖尿病肾病肾小球硬化,发挥其肾保护作用。更多还原

【Abstract】 AIM To investigate the effects of Tongluo Yishen Formula on the proliferation,expression of extracellular matrix control system of MMP-9/TIMP-1,transforming growth factor beta1(TGF-β1) of rat glomerular mesangial cell cultured in high glucose,and to explore its possible mechanism for preventing diabetic nephropathy progress.METHODS Rat glomerular mesangial cell cultured in high glucose was divided into normal control group,high glucose group,Tongluo Yishen high,medium,low dose groups and western medicine control group.Each group deliveryed with rat serum containing drug for 48 h.Cell proliferation was detected by methyl thiazolyl tetrazolium(MTT),and cell culture supernatants of TGF-β1,MMP-9 and TIMP-1 contents were measured by ELISA.RESULTS Compared with the normal control group,the glomerular mesangial cell proliferation,TGF-β1,TIMP-1 expressions significantly increased and MMP-9 content decreased in the high glucose group(all P<0.01).Tongluo Yishen high,medium and low dose groups all significantly inhibited glomerular mesangial cell proliferation(P<0.01 or P<0.05),lowered the expression of TGF-β1 and TIMP-1,and raised MMP-9 expression(P<0.05 or P<0.01).Those in high dose group were the best in dose-dependent manner.There were no significant differences of all these indicators between western medicine control group and Tongluo Yishen medium dose group(P>0.05).CONCLUSION Tongluo Yishen Formula can antagonize diabetic nephropathy glomerulosclerosis,and its renal protection may be related to inhibiting glomerular mesangial cell proliferation,downregulation of TGF-β1 and promoting the degradation of extracellular matrix.更多还原
