黑酵母β1,3-1,6葡聚糖对大鼠全身电离辐射损伤的保护作用

【摘要】 目的观察黑酵母β1,3-1,6葡聚糖对大鼠全身辐射损伤的保护作用。方法将35只SD大鼠随机分为6组:正常对照组(c组),单纯给药组(m组),5 Gy单纯照射组(5 Gy组),5 Gy照射并给药组(5 Gy+m组),0.5 Gy单纯照射组(0.5 Gy组),0.5 Gy照射并给药组(0.5 Gy+m组)。各组均行自身对照。大鼠自照射前2周给予黑酵母β1,3-1,6葡聚糖灌胃[1 g·(100 g)-1,qd]。对大剂量辐射组,检测照射前、照射后3,7,14 d体质量、外周血常规、血清超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量及照射后14 d骨髓细胞微核率和脾脏指数;对小剂量辐射组,检测照射后2,4,8,24 h,外周血淋巴细胞DNA损伤免疫荧光焦点数。结果黑酵母β1,3-1,6葡聚糖可促进照射后大鼠体质量恢复,减轻骨髓抑制,延迟SOD活力降低和MDA升高,差异有统计学意义,降低微核率增加,差异有统计学意义,抑制脾脏缩小;对接受小剂量照射的大鼠表现出DNA断裂位点修复的作用。结论黑酵母β1,3-1,6葡聚糖对SD大鼠全身辐射损伤有一定的保护作用。

【Abstract】 Objective To observe the protective effect of black yeast β1,3-1,6 glucan in rats’ radiation injury. Methods Thirty-five SD rats were randomly divided into six groups: control( C group); simple medicine administration( m group); 5 Gy radiotherapy( 5 Gy group); 5 Gy radiotherapy with medicine( 5 Gy + m group); 0. 5 Gy radiotherapy( 0. 5 Gy group); 0. 5 Gy radiotherapy with medicine( 0. 5 Gy + m group). Before irradiation,all the rats were given with the black yeast β1,3-1,6 glucan for two weeks except C group,5 Gy group and 0. 5 Gy group [1 g·( 100 g)- 1by mouth,once daily]. For groups receiving high-dose radiation,the body weight,peripheral blood,serum SOD activity and MDA content before irradiation, and 3,7 and 14 days after irradiation were detected. The bone marrow cell micronucleus and spleen index on the 14th day after radiotherapy was also measured. For groups of low-dose radiation,DNA injury was evaluated with immunofluorescence focus number of peripheral blood lymphocytes 2,4,8 and 24 h after irradiation. Results The black yeastβ1,3-1,6 glucan is effective in promoting weight recovery,reducing bone marrow suppression,delaying of SOD activity reduction and increasing MDA content after irradiation( the difference was statistically significant). Black yeastβ1,3-1,6 glucan reduced the micronucleation rate,and the difference was statistically significant. It appeared to maintain the original size of the spleen to protect the immune system,and repair the DNA injury after low-dose of irradiation. Conclusion The black yeastβ1,3-1,6 glucan has some protective effect on SD rats with systemic radiation injury.
