氧化低密度脂蛋白诱导小鼠心脏内皮细胞炎性反应中Toll样受体2的表达
The expression of TLR2 receptor in cardiac endothelial cells isolated from mice with inflammatory response induced by ox-LDL
【摘要】 目的研究氧化低密度脂蛋白(ox-LDL)诱导小鼠心脏内皮细胞炎性反应中Toll样受体(TLR)2的表达。方法分离小鼠心脏内皮细胞,对照组给予DMEM培养,干预组分别给予不同浓度的ox-LDL(20,40μg/mL)作用细胞24 h,采用Western blot法检测细胞间黏附分子-1(ICAM-1)、TLR2的表达,利用ELISA方法检测炎症因子白细胞介素(IL)-6、IL-8的变化。结果与对照组相比,ox-LDL干预组TLR2蛋白表达减少(P<0.01),ICAM-1蛋白表达增多(P<0.01),IL-6和IL-8表达明显增加(P<0.05);与低浓度组比较,高低浓度组ICAM-1、IL-6和IL-8表达差异无统计学意义(P>0.05)。结论 ox-LDL可以诱导小鼠心脏内皮细胞的炎性反应,同时可能抑制了TLR2受体的表达。
【Abstract】 Objective To research the expression of TLR2 receptor in cardiac endothelial cells isolated from mice with inflammatory response induced by ox-LDL. Methods Cardiac endothelial cells were isolated from mice, ox-LDL(20, 40 μg/mL) was added into the medium for 24 h retrospectively in the treatment group, while only DMEM was added in the control group. Chemokine ICAM-1 and TLR2 receptor was detected by Western blot, cytokine IL-6 and IL-8 were determined by ELISA. Results Compared with the control group, ICAM-1, IL-6 and IL-8 increased in the ox-LDL treatment group(P < 0.05 or P < 0.01), while TLR2 receptor decreased in the ox-LDL treatment group(P < 0.01). There were no differences between low and high concentration group in the expression of ICAM-1, IL-6 and IL-8(P > 0.05). Conclusion ox-LDL can induce inflammatory response in cardiac endothelial cells isolated from mice, while may inhibit the expression of TLR2 in cells.
