电针对阿尔茨海默病模型大鼠海马胆碱能神经元的影响

【摘要】 目的观察电针对阿尔茨海默病(Alzheimer’s disease,AD)大鼠海马乙酰胆碱酯酶(AchE)和乙酰胆碱转移酶(CHAT)的影响。方法通过Morris水迷宫筛选48只健康Wistar大鼠随机分为4组。除正常组外,其余3组大鼠均注射β-淀粉样肽(1-40)(Aβ1-40)所致AD模型。模型组不治疗。加兰他敏组采用氢溴酸加兰他敏灌胃。电针组造模后选取百会、双肾俞、双足三里、大椎等穴位针刺治疗。治疗4周后各组大鼠处死取海马,用免疫组化法和考马斯亮兰法测定AchE、CHAT的表达和含量。结果模型组AchE每个视野阳性细胞表达明显增强并且含量[(348.2593±13.0672)IU/g]升高,与正常组、加兰他敏组和电针组比较差异有统计学意义(P<0.05);但模型组CHAT阳性表达减弱且含量低[(81.8543±4.6528)IU/g],治疗后加兰他敏组[(205.7539±5.9732)IU/g]和电针组增强[(202.5637±5.3695)IU/g),与模型组比较差异有统计学意义(P<0.05)。结论电针通过调节胆碱能神经元的表达,以促进受损神经的恢复和再生。

【Abstract】 Objective To study the effect of electro-acupuncture on AchE and CHAT in hippocampus of rats with Alzheimer’s disease.Methods 48 healthy Wistar rats were screened by the morris water maze,and divided into the normal group,model group,Galanthamine group and electro-acupuncture group randomly.In addition to the normal group,the rats of the other three groups were injected Aβ1-40 of AD model.The model group didn’t give any treatment.Rats in Galanthamine group were lavaged with Galnthamine hydrobromide.Rats in electro-acupuncture group select Baihui,double Shenyu,double Zusanli,Dazhui for acupuncture treatment.After 4 weeks,each group of rats were died and taken out of the hippocampus,the expression of rats in hippocampus AchE and CHAT were detected by immunohistochemical and coomassie brilliant blue method.Results The expression of rats in hippocampus AchE in the model group [(348.2593±13.0672)IU/g] was obviously improved,the difference was statistically significant(P < 0.05);compared with the model group [(81.8543±4.6528)IU/g],the expression of rats’ hippocampus CHAT in the Galnthamine group [(205.7539±5.9732) IU/g] and electric-acupuncture group(202.5637±5.3695 IU/g)was obviously improved,the difference was significant(P < 0.05).Conclusion Electro-acupuncture can regulate cholinergic neurons expression in order to promote the recovery and regeneration of damaged nerves.
