脂氧素A4对心肌缺血再灌注损伤大鼠细胞因子和氧化应激的影响
Influence of LipoxinA4 on Cytokine and Oxidative Stress in Rats Undergoing Myocardial Ischemia-reperfusion Injury
【摘要】 目的探讨脂氧素A4(LXA4)对心肌缺血再灌注损伤(MIRI)大鼠细胞因子和氧化应激的影响,明确其心肌保护作用。方法 72只雄性SD大鼠随机分成假手术一组(C1组)、假手术二组(C2组)、MIRI一组(I/R1组)、MIRI二组(I/R2组)、MI-RI前用药组(LX1组)、MIRI后用药组(LX2组),每组12只。建立大鼠MIRI模型,各组于开胸前取血(T1)、实验结束后取血(T2)测IL-1β、IL-6、IL-8、IL-10、TNF-α、cTnI血清浓度,同时测定MPO、SOD活性、MDA含量及病理切片观察心肌炎性细胞浸润。结果 I/R1、LX1与C1组相比,I/R2、LX2与C2组相比,血清IL-1β、IL-6、IL-8、IL-10、TNF-α、cTnI浓度(均为T2),MPO、SOD活性和MDA含量显著增高;LX1组与I/R1组相比,LX2组与I/R2组相比,能显著降低血清IL-1β、IL-6、IL-8、TNF-α、cTnI浓度(均为T2)及MPO活性、MDA含量,并显著提高血清IL-10浓度(T2),SOD活性(P<0.01)。MIRI各组心肌炎性细胞浸润明显增多,而LX各组心肌炎性细胞浸润明显减少。结论在MIRI前、后应用LXA4均能抑制中性粒细胞浸润和活化,降低促炎细胞因子释放,促进抑炎细胞因子分泌,减轻组织氧化应激,对大鼠MIRI起明显的保护作用。
【Abstract】 Objective To investigate the influence of lipoxinA4(LXA4) on cytokine and oxidative stress in rats undergoing myocardial ischemia-reperfusion injury(MIRI) to confirm the myocardial protective effects of LXA4.Methods Seventy-two male SD rats were randomly divided into six groups,with 12 in each group:sham operation group one(group C1),sham operation group two(group C2),MIRI group one(group I/R1),MIRI group two(group I/R2),pre-MIRI treatment group one(group LX1),post-MIRI treatment group two(group LX2).The rat model of MIRI was established.Blood was taken in each group before chest opening(T1) and at the end of the experiment(T2).The serum IL-1β,IL-6,IL-8,IL-10,TNF-α,cTnI concentrations were measured and at the same time the MPO,SOD activity and MDA content were checked while the pathological myocardial inflammatory cell infiltration was also observed.Results In group I/R1,LX1(compared with group C1) and group I/R2,LX2(compared with group C2),the serum IL-1β,IL-6,IL-8,IL-10,TNF-α,cTnI concentrations(all at T2),MPO,SOD activity and MDA content increased significantly.In group LX1(compared with group I/R1) and group LX2(compared with group I/R2),lipoxinA4 could significantly reduce the serum IL-1β,IL-6,IL-8,TNF-α,cTnI concentrations(all at T2),the activity of MPO and MDA content,at the same time,could increase the serum concentration of IL-10(T2) and SOD activity(P<0.01).Inflammatory cell infiltration in MIRI groups increased significantly,while it was significantly reduced in LX groups.Conclusion Before and after MIRI,application of LXA4 can inhibit neutrophil infiltration and activation,reduce the release of pro-inflammatory cytokines,promote suppressive inflammatory cytokines secretion,and alleviate the tissue oxidative stress.LXA4 has protective effects obviously against MIRI in rats.
