辅助生殖技术中废弃胚胎继续培养的影响因素分析

【摘要】 目的探讨辅助生殖技术中废弃胚胎继续培养的潜能,为今后利用废弃胚胎建立胚胎干细胞系提供参考。方法将D1观察为无原核(0PN)、单原核(1PN)、多原核(≥3PN)和受精正常(2PN),D3评分为Ⅳ级的胚胎继续培养至囊胚期,观察囊胚形成率。比较年龄、受精方式、不同来源、D3卵裂球细胞数、D3胚胎碎片与囊胚形成率的关系。结果共收集143个取卵周期499个废弃胚胎进行继续培养,形成囊胚105个(21.04%),优质囊胚为24个(22.86%)。年龄、不同来源、D3卵裂球的奇偶性、D2卵裂球的数目各组之间差异不显著(P>0.05);受精方式、D3卵裂球数目、胚胎碎片各组之间差异显著(P<0.05)。结论部分废弃胚胎有继续发育成囊胚的潜能,受精方式、D3卵裂球的数目和胚胎碎片情况可能为囊胚形成的主要影响因素。

【Abstract】 Objective To explore the developmental potential of discarded embryos in assisted reproductive technology,thus to provide the reference to set up the embryonic stem cell lines in future.Methods All embryos with non-pronucleus(0PN),a single pronucleus(1PN),a number of pronucleus(3PN) and normal fertilization(2PN) but score for Ⅳ level embryos in d3 were cultured into blastula by the sequential method.The blastulation rates were observed.The relationships between the blastula formation and the patient age,fertilization way,embryos from different sources,the blastomere numbers and fragments at d3 were studied.Results A total of 143 fresh cycles about 499 discarded embryos were collected and formed 105 blastulas(21.04%),among which 24 were high-quality blastulas(22.86%).The patient age,embryos from different sources,the parity of the blastomere number and the blastomere number at d2 in each groups were no significant differences(P>0.05).The fertilization way,the blastomere number at d3,fragments in each groups were significant differences(P<0.05).Conclusion The discarded embryos have development potential on different degrees.The fertilization way,the blastomere number at d3 and the fragments were the main effect factors of culture to blastocyst.
