白藜芦醇对异氟醚吸入诱发老年小鼠认知功能障碍的影响

【摘要】 目的研究白藜芦醇对异氟醚吸入诱发老年小鼠认知功能障碍及海马组织炎症反应的影响。方法 48只15个月龄雄性C57BL/6小鼠随机均分为四组:A和B组连续7d腹腔注射生理盐水后分别吸入氧气和1.5%异氟醚2h;C和D组连续7d腹腔注射白藜芦醇100mg/kg后吸入氧气和1.5%异氟醚2h。气体吸入后24h行旷场实验和条件恐惧性实验;6h和48h取海马组织,采用ELISA或Western blot法检测IL-1β、TNF-α、NF-κB抑制蛋白α(IκBα)和Nod样受体家族包含pyrin结构域蛋白(NLRP3)水平。结果 B组环境诱发僵直时间百分比低于A、D组(P<0.05)。气体吸入后6h和48h,B组海马IL-1β、TNF-α和NLRP3水平高于A、D组(P<0.05),IκBα水平低于A、D组(P<0.05)。结论白藜芦醇可改善异氟醚吸入诱发的小鼠认知功能障碍,可能与其减轻海马组织炎症反应有关。

【Abstract】 Objective To study the effects of resveratrol on the isoflurane-induced cognitive impairment and the inflammatory responses in the hippocampus in aged mice.Methods Forty-eight15-month male C57BL/6 mice were equally randomized into four groups.After injected intraperitoneally with saline daily for 7consecutive days,the rats in groups of A and B were inhaled oxygen and 1.5%isoflurane,respectively,for 2hours.After injected intraperitoneally with resveratrol100mg/kg daily for 7consecutive days,the rats in groups of C and D were inhaled oxygen and 1.5%isoflurane,respectively,for 2hours.The mice were tested with the open field and fear conditioning at24hafter gas inhalation.The hippocampus was harvested at 6hand 48hafter gas inhalation for the determinations of IL-1β,TNF-α,IκBαand NLRP3by ELISA or Western blot.Results The percentage of the contextual conditional freezing time was lower in group B than that in groups of A and D(P<0.05).The IL-1β,TNF-αand NLRP3were higher,but IκBαwas lower,in group B than those in groups of A and D in the hippocampus at 6hand 48hafter gas inhalation(P<0.05).Conclusion Resveratrol attenuates the isoflurane-induced cognitive impairment in the aged mice,possibly attributing to the attenuation of the inflammatory responses in the hippocampus.
