他莫昔芬在大鼠蛛网膜下腔出血后早期脑损伤中的神经保护作用

【摘要】 目的探讨他莫昔芬对大鼠蛛网膜下腔出血(SAH)后早期脑损伤的神经保护作用。方法 60只健康成年雄性SD大鼠随机均分为四组:对照(BC)组、SAH(MC)组、安慰剂(VC)组和他莫昔芬治疗(TT)组。应用视交叉前池一次注血法制成SAH后早期脑损伤模型,在SAH后2、24及36h分别腹腔注射他莫昔芬;48h后处死大鼠,取颞底皮层脑组织做标本,测定脑水肿指数及血脑屏障变化。结果与BC组相比,MC组大鼠脑组织含水量明显增加,血脑屏障破坏明显;与MC组相比,TT组脑水肿指数降低,血脑屏障功能改善。结论他莫昔芬能有效降低SAH后脑水肿、改善血脑屏障,提示他莫昔芬对大鼠SAH后早期脑损伤具有神经保护作用。

【Abstract】 Objective To investigate the neuroprotective effect of tamoxifen in early brain injury in rat model with subarachnoid hemorrhage(SAH).Methods Sixty health male SD rats were egually randomized into four groups of BC(blank control),MC(SAH model control),VC(vehicle control)and TT(injecting tamoxifen 5mg/kg intracisternally at 2,24and 36hafter arterial blood injection).SAH models in groups of MC,VC and TT were established by injection of 0.3ml fresh blood into prechiasmatic cistern in 20seconds.Cerebral edema index and blood-brain barrier(BBB) permeability were detected at 48hafter SAH.Results Compared to group BC,brain water index was increased and BBB damage was increased in group MC.Compared to group MC,brain water index in the cortex was decreased and BBB was improved in group TT.Conclusion Tamoxifen treatment can significantly ameliorate the brain edema and BBB impairment in rat model with SAH,suggesting that tamoxifen has neuroprotective effect in early brain injury in SAH rats.
